Not sure how to proceed with this grow

Hello friends

I've got some plates that I started about 10 days ago with some "Argentina" spore solution. This is my first time growing and I'm using some ziploc containers as plates and plan to use this Broke Boi tek.  Here's what the plates look like:

They've all got what looks like some proper healthy mycelium growing, along with maybe some unwanted guests.  8 has definite mold, 7 looks like it might have 2 different fungi growing, and they've all got spots that I think are bacterial.  I admit I could certainly do better with my technique, like investing in an inoculation loop instead of just squirting some spore solution on top of the agar. 

Where do I go from here?  I suspect 8 is a lost cause, but what about the rest? Can I use them as is to spawn to bulk?  Do I need to transfer to new plates?  Can I use any of this at all?

Any advice would be much appreciated.

I've got another 8 containers on deck I can put more agar in.

So do I just cut a little wedge off outer edge of each mycelium growth and set it in the middle of the new plate?

I think what would work best is if you take a dive into the agar envy thread and take a look at what healthy mycelium looks like and then from there suggest what part of which plates you think would be good contenders for transfers. Seems like a good learning practice.

For transfers (and all things agar related really) you only need the smallest section to move to the next plate (or inoculate grain). Since you are still running away from contamination you will want to take a section from the leading edge of growth. When you have a healthy plate you can take transfers from any section theoretically.

I think green is not the best color for agar work.

I would go with the bottom colony on 7 personally. Looks like it would be clean fastest. A few of them could be good already but you never know with such a big leading edge.

Do you have a flow hood? or did you take those pics inside a SAB?

You def want to wait until you have clean plates before transferring to grain, those all look suspect even at the outer edges except maybe on the bottom colony of 7, though that looks really thick and kinda strange too.

I would take a couple little wedges from a few different areas on a couple of them and just see how they grow out. I always recommend petris for people new to agar because they're much easier to work with cleanly. With these you have to tilt them basically to be able to cut a wedge without hovering over them and then the lid will either have to be set down or held somehow in your off hand

I'm seeing recommendations on other threads to dunk wedges in H202 for a few minutes before transferring. Is this a way to kill bacteria on the wedge?

When you say "big leading edge", do you mean I let the plates grow out too long?  Should I have transferred earlier?

It was inside a plastic tub I'm using as a SAB.  I didn't cut holes out of the sides though.  I just let it sit over the edge of a table and stick my arms up under.  It's all sitting on a lid that I sanitize with isopropyl before using, though I understand that doesn't make it sterile.  Would it help to tape the lid to the container on one side to keep it more manageable, or would that be a potential source of contam?

No I mean the big line of clear growth surrounding the myc. Sometimes that can be mold hiding.

I let my plates grow to the walls all the time as long as they’re clean.

I looked at that broke boy tek and I must say that I don’t like the idea of steaming whole brown rice for only 90 min. That sounds ridiculous. He has no pics of the results, that’s a bit red flag. If the results don’t look like something I want for myself, I’m not going to wast the time with it.

Good results look like this IMO

I have an instant pot so I'll be using that to sterilize the rice jars. I already have a big sack of brown rice an old roommate left behind that I have no intention of eating, so this tek seemed like a good way to put it all to good use. 


As for what parts to transfer, folks seem most confident about the bottom of 7 so I'll focus on that.  Looks like maybe the top right of #1 and bottom right of #4 could also be saved.  The rest I think I'll grow out for another week separate from the new plates to see what happens with them.

EDIT:  It just occurred to me that I did not wear a mask while doing any of this work, which seems a real stupid oversight on my part.  Whoops. 

EDIT 2: What do you actually do with a clean plate of healthy myc?  Does all that that need to be transferred and grown on more plates, or is one plate  enough to multiple jars for one grow?
