Syringe to Agar

Hey all,

Just started working with agar. Been using prints mostly. I did have a few cc's left in a syringe today and did my best to squeeze a single drop onto a plate. I ended up squirting it all over the place lol. I quickly capped it but I reckon most of the spores are gone to the side of the dish. If the mycelium does start growing on the side what is the best course of action? Would you just go plate to plate before full colonization there by starting growth from the middle? I don't want it growing through the parafilm.

Using a syringe to agar for the first time can be messy. It was for me. That's what transfers are for

You don't NEED to inoculate it right in the middle of the plate. It would definitely be better to not have it right on the edge, but no matter where you inoculate it should grow radially outwards and you could find sectors that could be used for grains, LC, etc.

For future reference, after flaming your needle you can cool it off by forcing a few drops of solution onto the floor of your SAB. One you’ve done that, the tiniest amount of pressure will cause a single drop to fall.

Yea Psilicon is totally right.

If you flame a syringe and then hop on over to your plate and try to squirt it, there will be a lot of resistance and sudden release.

Instead, flame, squirt a bit somewhere to cool and loosen up the pressure, then move to your stack of plates and go one at a time barely pressing and it is really easy to get a single tiny drop to go. So you dont flame in between each single plate, just before you shoot your stack of however many in one motion.

For some reason the very first movement of the plunger can require a fair amount of force and can end up squirting out more solution than required, so if you get it out of the way in your SAB first after flaming the needle, it will not only make it easier to push out a single drop when needed, it also helps to cool then needle.  Get two birds stone at once!

syringe to agar sucks and anyone whos ever done it hates it. but heres why i do and this is a good example of why we use agar. this syringe came from if you ask me the best vendor there is period and it shows that you should never just shoot shit into grains. yes it can work but it will often lead to some kind of contamination usually bacterial in my experience.

I keep sterile swabs handy. Once I get a droplet on the agar surface I use the swab to streak it in a distinct line or zig zag pattern. It help to keep from having liquid rolling around the plate.

Yea syringe to agar sucks but I mean it's like the only thing you can safely do with a syringe. It's more like syringes just suck...

If it was going to be dirty on agar it was going to be dirty on grain or LC so ...

It is a slight pain to squirt that drop and much easier to use prints and a swab for agar for sure.

Syringes are lame.

I also keep sterile swabs handy and I also do essentially the same thing as you, the only difference is that I drop one drop directly onto the swab, then streak it across the plate. Zero excess water.
Two sides of the same coin though.

Couldn't agree more. Yes sometimes it will work, but more times than not you will end up with bacterial contamination.

I have syringes right now that are so bacterial, not only will they not germinate on agar, 9 times out of 10 they won't even germinate on grain!
When this is the case, I'll make a dozen small jars of grain and just pray to god that at least ONE of those jars will at least get a tiny bit of mycelial growth,  then I can pluck one grain and put it to agar, and start cleaning from there. 
I have a couple cultures like that in the works now.  Couple more transfers and they should be clean.

This thread has helped me feel better immensely. I have very little experience in this field and I have been having terrible results with syringe to agar over the past 2 months. But at least now I feel like less of an asshole :tongue: :thumbup:

I now have a better method than what I stated above to germinate dirty syringes.
I just make small BRF cakes using my agar dishes and shoot a drop of spore solution on them. BRF seems to be far more resilient to bacteria than agar or grain.  I've never had a BRF dish like this not germinate, no matter how bacterial the syringe.
Once you see clean growth, you can transfer a piece to regular agar and continue cleaning like that, or you can even let the plate cake fully colonize and fruit it, or just pluck a pin and clone that.

You're essentially just doing the PF tek (only on a smaller scale) and cloning or transferring myc from there.
