Colony selection from germ plate

When streaking spores to agar, how should the initial transfers be selected? I had a germ plate that grew well. All of the colonies looked very similar. I transferred three clean samples to different plates, but they seemed very sluggish. I looked back at the germ plate, 6 weeks old at this point, and saw there were some sections with rhizo growth and even some knots. I took new samples from these sections and these new plates are definitely more vigorous.

Is there something in particular that I should be looking for in early growth, or should I be waiting to see some different phenotypes in the germ plate before I transfer?

You just want to transfer clean, healthy looking germination that is as far away from any bacteria or mold colonies on the plate as you can get.
There are going to be many, many different strains present in the myc you end up transferring anyways.

I like to streak with a loop, it makes it easier to identify myc germination - I will definitely choose growth in the streak pattern than something off somewhere else on the plate - mold colonies often look just like germination

I concur with the punx, and in general you cant tell if a colony is going to have desirable qualities from germ, so you pick the cleanest, nicest looking ones on the streak pattern.
Its one of the few parallels in growing plants, you cant tell if a specific young seedling is going to be viable or healthy fruit well, but if its germinated and grown quickly, thats a good sign.
