I've been having this issue lately and I thought my potato flakes had gone bad or something, I think I just haven't been getting the mix hot enough. I've had agar break apart it chunks rather then a gelatin consistency even after PCing if that makes any sense.

Use a different agar recipe the one in this Tek is literally trash.

@rudimentary I've never done potato agar and tbh I never will if I can, it just sounds ghetto to me. Get your hands on some malt extract, or alternatively try grain water/grain flour agar.

I have had success with this but using BRF and actually 0 dextrose or honey. I read about brf on some other tek, so maybe try that. I couldnt find potato flakes and bought powder instead, but then I realized the brf is higher nutes. Transferred mycelium doing really well. Growing super fast.

I've experimented with all sorts of ways with this and I have had success with original recipe as well. I think it just boils down to genetics for example I made a bunch using the original recipe and used different strains on different plates. Some germinated fast and some took forever

I recently had an issue with my no-pour plates. I sterilized a batch, but I didn't use them for about a week (might've used just one, don't remember) and left them sitting on a shelf. I did not use paper towels because I didn't have any at the time. I never took off the foil. After some time they started growing molds - every single one of them, and they were never opened. I sterilized for 30 minutes, I believe. I saw 30 minutes for mini rounds on a different tek. Was that my problem? That seems really strange to me. I'm scared to make more plates because I'll be germinating spores on them, and I don't want them to just contaminate. Fucking everything I've grown for the last few months has contaminated, it's infuriating. That's why I'm back to spores, cause all my cultures are fucked.

If you didn't open them and they contamed it means you didn't sterilize properly. Did you vent your PC?

Vent for 10 minutes? Yeah, I always do that. I mean, sometimes I might vent for like 8. I did actually have a little leak in my gasket, but I took it out before that cycle and cleaned it and it seemed to be okay? I think there was debris in there.
Honestly I think I'm a little too afraid of caramelizing my media and I'm always riding my stove heat to keep it right. It's so sensitive though, and sometimes it drops below 15. Maybe that's how I fucked up.

Is light malt extract the preferred one? I saw some malt extract at the grocery store but I don't think it was light and I think its the only one they had.

If your agar is chunky after solidifying it's most likely either your brand of agaror you are evaporating too much water and not replacing it.

If I'm supposed to heat up my mix hotter than the tek says should I also add a bit more water to make up for what evaporates?

Itactualy says to boil it in the tek,then to pour it into a measuring cup and add back hot water to original amount

So sorry if this has been answered I tried finding what I was looking for but after looking through like 50 pages of the 364 I figured I might as well ask.
I made up an 8 pack of plates and intend to take them out of the PC today. I want to use two of them for a test and have 6 for now.
With unused pasty plates and also colonizing ones, what is the best way to store them? Can I leave them out in room temp and they will still last awhile? I'm looking to use them within maybe 2-3 months as I don't have alot of space for many tests at the moment. I have seen fridge is an answer given but didn't get alot of information about that. How long would they last out in the open as opposed to fridge? And wouldn't that be prone to contams since a fridge is not exactly clean?

I've stored fully colonized plates for weeks at room temp with no problems eventually the mycelium gets weak or dries out though, the fridge is good for longish type storage fully colonized plates aren't really at risk for contams until it gets old you also have a filter on top to stop contams if you're really worried cover it with foil or plastic wrap for long term storage. Contams have a hard time germinating and spreading in cold tenps. So you're good I just wouldnt keep a fully colonized plate at room temp for over a month.

If you need to store a culture on a plate for awhile its wise to pour thick and seal it up well.
