LC Questions

Well I did it again, got a good grow going and then neglected it to the point of basically starting over. I took one of the last fruits I got and started my first attempt at cloning. Lo and behold, success!



So, I'm 100% sure this plate is clean, but would it be worth it to do another transfer since this straight from the fruit? I don't know if there's any extra inherent risk going from first clone transfer straight to LC.

If that's not a problem, then on to the LC questions!

I'm planning to use some of my grain soak water in a media bottle. Adding some broken glass to it to agitate the culture then PC'ing just like agar. I've seen a lot of posts about needing a filter, but can I get away with this just loosening the lid for the PC process and retighten it to keep it clean during colonization? I'm under the impression that gas exchange isn't absolutely necessary during the colonization phase. Please correct me if I'm wrong!

Next question is how much of my clean culture will I need to add to the broth? Same as I would with agar transfers or the entire plate cut up into multiple pieces?

Clean growth is clean growth no reason you couldn't go from the original clone plate to an LC.

You could do what you are saying with the bottle and no filter.

A slightly larger size transfer that you would do to a plate is what I do to start a LC.
