Hi. I am new member here and greetings to all the members. Recently I purchased some cubensis spores (ecuador) and set up everything to make them grow (I was using brown rice flour method). After 10 days there are no visible signs of colonization on all 6 jars - except one (picture in attachment) where I did find some mycelium. I tried to do everything sterile, even made my SAB. I got a good print and used half of it for 6 jars (325 ml jars).
Don't know why they didnt grow (no sign of contamination), but here are my thoughts:
- I couldn't get jars with lids that have rubber circle on the contact with jar, so while I was sterilizing jars (not with pressure cooker but 1.5 hours in a big pot with closed lid) maybe some moisture got in the jars? I drilled 4 holes in lids and covered them with micropore tape.
- I used electricity stove so when I got it to boil, I put the setting on 1 (out of 3) but then I didnt hear any boiling so I turned it to 2 (slow boiling - I heard bubbles). So maybe I overcooked my vermiculite/BRF mixture? I used 1:1:2 ratio (water:BRF:vermiculite) in which I added slightly more water since I thought mixture is too dry. So it is basically 1.2:1:2 ratio. I added a little bit of gypsum as well.
- can spores be dead (they were travelling for one month to me and after they got here, I used it after a week)?
Please, if you have any advice based on the pics. I don't know where it got wrong:(

I'm still learning here but that pic looks like it might be mold as opposed to mycelium. Is it showing up directly below an inoculation hole?
If you only steamed your jars for 1.5 hours, I don't think you could have overcooked it, regardless of stove settings. Your substrate does look a little wet and clumpy though; if you're doing PF Tek you should try not to pack it down so as to leave it airy, but also you add dry vermiculite as the top 1/2 inch of so all the way to the top. Wouldn't recommend shaking the jars, but even if you were to shake there shouldn't be enough room to clump the substrate up that way.
That said, it's possible your spores were no good if you got no mycelium in 5 out of 6 jars and what definitely might not be mycelium in the 6th. Try one of the sponsored vendors.
"I got a good print"... did you buy the spores as a print and create your own spore syringe, or did you buy a spore syringe?

Got foil over the lids? How much cc of spores did you use per jar? Perhaps it got hardened from too much gypsum, which you really don`t need here.

I don’t do cakes so I’m not sure why but your mixture looks off. Probably too wet.

Hi. I followed all procedures as in Let's grow mushrooms video (same as the one on the link you provided). I got spore print from freespore.es site. I think my mixture is too wet because there are no rubber on the lids of my jar so during sterilization some could get in. Btw I used tight pice of foil on every jar during sterilization.
I made my own syringe. Used half print and got 20 ml of spore/water solution, which I tried to spread evenly on 6 jars (4 holes each jar). During inoculation of my last 2 jars I noticed that I had to press syringe with some more force, like something stuck inside the needle (never mind that, at least 1 jar should have succeeded). Does thickness of the needle matters? I have regular one used in medicine, with sharp edge...

So some of you think white stuff is mold? Shouldn't mold be grey in color, not white?

Depends on the kind of mold. Greyish would be mold like cobeweb. Trich starts out shiny white (brighter than myc), and becomes colour after sporulating.
I don`t think it`s due to the "no rubber" on the lids, you just want steam getting in, no boiling or dripping water. Most people here use the rubber side up for better handling in the SAB and there`s no issues.
But you may get some better jars for your own peace of mind
