First Agar (in many years) Tell me what you see!

Hey all! I just did up my first Agar plates in a while and I want to get some opinions on the pics. I've been struggling with Trich in my bins and I'm hoping to trace it back to the spawn as I've tried everything I can think of regarding cleaning the room/bins/SAB/pasteurizing CVG etc

I dropped some spores from my first ever spore syringe of my first ever spore print

expecting a bunch of contams and it seems like they just grew fine (beginner's luck?) Or I'm just not seeing the contams.

Transferred some wedges from each of the growths to new plates



Wondering about the whispy growth on this one. It looks like myc strands but thin. Does cobweb grow like this on agar?



Am I on track? Do i transfer from the edges of each of these? Am I supposed to be using an inoculation loop rather than cutting wedges? Can you see any contams that I cant?

You are on track, tomentose is totally normal on top for me and rhizo underneath, neither of them is a big deal.  You should just let those grow out a little bit for a couple of days before cutting for transfer in my opinion.

Okay well I transferred out the wispy stuff and just got a big ball of wispy stuff on the new plate, so I'm assuming that's a contam of some sort, or the world's weakest myc.

None of the rest look particularly healthy or agressive. Is that normal from spores? Do they need a while to mature before that rhizo growth comes along?

Also I'm having problems with condensation on the plates. How should the blanks be stored? They looked good until I put them into the fridge...

everyone gets condensation- i just tilt it and dump it out in the SAB when I take it out of the fridge to use  :shrug:

Just store them at room temp, somewhere without big temp swings if possible. And make sure to let the agar cool to around 117-120f before pouring your plates.

When spores first germinate you have tons of different strains forming and it's all chaotic, after a transfer it should get better, and after a couple transfers you should have real organized growth. If your agar is very nutritious you won't see rhizo growth, and some cultures are just tomentose.

Those plates all look a little funny for some reason. Some times it's hard to tell what's up in pictures. Are these APE by chance?

Nope they're GT from a print/syringe I made myself.

I agree they all look funny. I'm wondering if it's because it was a 5+yo premade agar I bought, and had turned to stone. I powderized it and it seemed to work well, stuff is growing, but I wonder if something in it ages poorly.

My newest plates are fresh Agar and light malt extract, I'll see how they grow on there. Making a few transfers today and I'll update in a few days!

Ok cool, I'd like to see them on the newer plates because the agar itself can definitely effect growth and looks.

Alright so I did a few transfers and, well, oof. They don't look so good.

Also most of my plates have heavy condensation on top which doesn't seem to be clearing up. My mistake putting them in the fridge I guess. So it's hard to take pics but I can usually find an angle to see clearly.


Here's some spores surrounded by what I'm sure is bacteria




Some green in the middle. Maybe I can transfer away from it on the next plate?
