Sterile lab checklist

If one theoretically had a decent budget and about 500 sfqt with 2 small offices, what would your lab look like?
Would these be the "must haves":
Laminar flow hood desktop 20l autoclave (sterilization of tools, LCs, etc) standing autoclave 100l (grains) fridge with windowed door
What else should one consider?
Thanks!

Check out Meyer's Mushrooms, he's always posting videos about the machines he builds or adapts. Especially would recommend looking at his system for filling grain bags and adapting a broken dryer to make a spawn bag tumbler. Those two could save you a ton of time! If you have any questions you can talk to him directly, he's very helpful towards DIYers. Also the folks over in Gourmet and Medicinals sometimes have these commercial sort of machines.
Good luck!

Also is there a preference over vertical vs horizontal laminar flow hoods?

What you have is the essentials besides "burnables" (things I burn through) like agar, petris, etc.
As for other things I would want but dont necessarily need:
I would get some nice stainless steel lab tables on casters. A few wire shelving units to hold jars and bags etc. Build a steamer for sawdust blocks if your intention is to do edibles at all. Impulse sealer for bags Cement mixer. I use one to mix up sawdust subs and they work amazingly.
Comfortable rolling lab chair. This is a must, yo!!!

A few essential non disposables.. lighting and a decent magnetic stirrer for LCs. Maybe a temp gun (thermometer) to help with agar pour.

Quote:
Choose a horizontal laminar flow hood if:
-Your application requires less turbulence on the work surface - While both horizontal and vertical produce very little turbulence, horizontal may render slightly less as the airflow does not hit any obstruction until exiting the fume hood (as opposed to the vertical flow hitting the bottom of the unit). -You will be using primarily small utensils and equipment that will not cause airflow disturbance -Large equipment within the path of horizontal airflow may cause greater turbulence. -You need the most control over contamination. Hands and gloves are positioned downstream of the sample in a horizontal flow, therefore greater contamination protection is possible with horizontal laminar flow.
Choose a vertical laminar flow hood if:
-Your application necessitates large equipment be used on the work surface - Vertical flow does not produce as much turbulence when hitting large items within the flow, as opposed to the disturbance created in a horizontal flow. -Your application requires that soldering fumes or fine powders be used - Soldering vapors can be toxic due to the presence of metal oxides and acidic fumes from flux. Only use laminar flow hoods for handling non-hazardous powders or nuisance odors being generated on the work surface. -You need a taller, larger work space - Because the filters are generally positioned on top of a vertical laminar flow hood, there is greater height available for work surface.

Great snippet El Chupa, thank you. So for mycology a horizontal is likely the better option. Mainly for agar work and grain bag inoculation. The grain bags are the medium/large 6lb so placing a bunch in the flow hood and cabinet may cause a lot if turbulence?

I'll preface this with, I'm still learning a lot and my work history is in factories and self employment lol. With that said I believe that really anything in front of the flow causes turbulence but that's not a worry as long as your technique is solid. Anything that is being opened and worked on is farthest upstream and the turbulence it creates is downstream. If I'm mistaken in anyway then it'll be a learning lesson for me as well.

Yes i figured to placing work flow starting in the front of the hood make rotating forward as you go along makes the most sense. Thanks!

I'd start off with a PC, SAB, and the essentials for agar work initially until you know it's something you really enjoy. All of the equipment you have listed (assuming it's quality equipment) will run in the thousands altogether. But you can get started with a couple hundred easily. If you already have a good bit of experience with mycology and know it's something you wanna dump a good bit of cash in, then ignore me.

Hi Guys. I do have some experience with SAB, sterile tek, etc. I just want to take it to the next level with ease of use and convenience. I am, as always, still learning. I do appreciate the responses so far. We are looking to scale with gourmet edibles as well so i figured I better get comfortable in a lab. I have a small budget of about $5-7k.

Lol. But realistic. My lab for 80% of my mushroom growing was a living room table and plastic tub. Some gloves and rubbing alcohol.
You know people who need to buy the same running shoes Olympic athletes use before trying running out. Vs I go for a run in jeans and work shoes people.

They have water baths that maintain a good temp for agar to always be ready to pour. But a lab grade one can be pretty expensive. A decent, cheaper alternative is a sous vide

Listen i get it, you can do everything at home in a SAB and sterile tek.
My particular situation is a little different. I have investors looking into the medicinal and gourmet market which I will be assisting them with. They will be funding the lab for my work. I have 500sqft office to build. I need to get a shopping list together by Monday
Thanks everyone for participating in this thread, it will be really helpful.
On my wishlist so far:
1. large capacity steam autoclave 150l-360l 2. desktop autoclave 24l-80l 3. 2 person laminar flow hood and cabinet with horizontal flow, UV sterilization light, H14 HEPA 4. water bath/sous vide 5. fridge with window 6. stainless tables on castors 7. wire shelving 8. magnetic stirrers x 4 9. Maybe a bead sterilizer for tools 10. comfy chair
Misc lab supplies:
1. petri dishes 2. media bottles 3. maybe a microscope
What else??

I'm tellin ya a cement mixer is seriously awesome for mixing up sawdust substrate.
After a few times mixing by hand you'll regret not having one.
What are you planning on using as sub actually? I guess if you're using straw or something you won't need a mixer.

We have been using a cement mixer attachment to a drill but have been looking into the standing drum mixer you are referring to as I have shredded 2 of them already lol. Using the typical CVG 1:1.
We are looking into contracting a local supply store to pre-pasturize our subs. i just dont know how long they will stay pasteurized however. (i also question if its even necessary as I have done a bunch with zero pasteurization with the same results)

Do it. Look on craigs and shit and you may find one for relatively cheap.
@bod-I've never used straw for anything. I do all my edibles on supp sawdust blocks, but if I did I'd do that for sure. Unless I was pumping out quantity, in which case I'd get a shredder
Straw just seems like such a pain in the ass. I've heard oysters perform better on straw but I dont run oysters much.

Just a heads up, all that listed is more than likely going to be way out of budget unless you go the used route. A steam autoclave that large and the desktop autoclave will pretty much take up the entire budget on their own. An AA75x (for the desktop autoclave) for example is roughly 39 liters and sits over $1000 after taxes and shipping. Just something to keep in mind as you budget.

Bod is a minimalist. I can appreciate a minimalist HOWEVER, I think there's an argument to be made for somebody who spends damn good money in this hobby to have what they feel are the appropriate tools and environment I know speaking from my own experience it forced me to not give up no matter my shortcomings and failures because it would be like burning a pile of cash. On the other hand I can see how a person could fuck themselves a little (or a lot) in the wallet. Seems like this could easily go in the financial advice forum or whatever it's called here on shroomary, if we're talking purely cultivation advice then fucking do it OP.

Small scale for a few years. Not a master mycologist by any means. I have been given an opportunity to expand however, which I would like to take advantage of. I want to make sure i am properly equipped. Been using PCs, SAB, etc. but with this budget I might as well do it right.
Scaling and proper equipment will enhance my learning process which I think is never a bad thing. Some may feel its overkill, perhaps, but if you want to take it to the next level...
Side question: is steam preferred over dry heat sterilization?

Absolutely steam sterilization is necessary. Dry heat sterilizing may work for solid material sterilizing like tools but steam is necessary for agar, sub, grain sterilizing

Lots of buckets. Lots of bits of cutlery. measuring jugs. A good scale or two (bigger kitchen scale for weighing mushies, subs and powders, smaller for agar and such). I'd kill for some steel shelving. Lots of totes to hold equipment and supplies. some bins for holding grain and such, I use a round rubbish bin and it holds around 30 kilos of grain.
