Yes it does. As long as your cap seals well, there shouldn't be a problem

Small scale for a few years. Not a master mycologist by any means. I have been given an opportunity to expand however, which I would like to take advantage of. I want to make sure i am properly equipped. Been using PCs, SAB, etc. but with this budget I might as well do it right.
Scaling and proper equipment will enhance my learning process which I think is never a bad thing. Some may feel its overkill, perhaps, but if you want to take it to the next level...
Side question: is steam preferred over dry heat sterilization?

Absolutely steam sterilization is necessary. Dry heat sterilizing may work for solid material sterilizing like tools but steam is necessary for agar, sub, grain sterilizing

Lots of buckets. Lots of bits of cutlery. measuring jugs. A good scale or two (bigger kitchen scale for weighing mushies, subs and powders, smaller for agar and such). I'd kill for some steel shelving. Lots of totes to hold equipment and supplies. some bins for holding grain and such, I use a round rubbish bin and it holds around 30 kilos of grain.

Rapjack said: Check out Meyer's Mushrooms, he's always posting videos about the machines he builds or adapts. Especially would recommend looking at his system for filling grain bags and adapting a broken dryer to make a spawn bag tumbler. Those two could save you a ton of time! If you have any questions you can talk to him directly, he's very helpful towards DIYers. Also the folks over in Gourmet and Medicinals sometimes have these commercial sort of machines.
Good luck!
This was the gem I’ve been looking for. Their video about the Bubba barrel will be absolutely game changing. Thank you for that info
