Cloning question...Agar
    #26827514 - 07/16/20 05:58 PM (3 years, 6 months ago)

Yesterday I cloned a fruit into two Petri dishes.  Just went to put them in a different spot and in one of the Petri dishes the clone moved.  I looked and it’s not completely touching the agar.  Just partially.  Should I have pressed it into the agar?  I just quickly opened the lid and put it in the center and closed it again.

Is this normal?  My other dish is fine.  It isn’t moving.  I’m not shaking it to test or anything though.  Lol

You're good. As long as it has some contact with the agar it'll be fine. Also, the smaller the area of contact, the less chance of contams also coming into contact with the media. For this reason, it is best to use as small of a piece as you can when cloning and doing agar to agar transfers.

Thanks for the advice.  They weren’t huge, maybe half an inch or so from the middle of the stem, but will go smaller next time now that you say that.  Makes sense.  The one has been in the same spot but the other bounced  around a little so we shall see.  Lol.  Hopefully my SAB game was on point.  Lol

It should be fine. If it is touching at all it will take to the agar without issue.
