Contams on pasty plates 3 weeks after innoc...

I've been growing cultures for several weeks now and I'm on my 4th set of past plates. The third set is showing signs of contams 3 weeks in while the second set didnt have hardly any before I trashed them. I dont want to start a master jar until I'm certain it's going to be contam free.
Started from spores on one, then a mycelium extraction the next 3.

Are you transferring the good myc onto fresh plates for the 4th time or starting from spores for the 4th time?

Fresh myc from the new plates. I cut out around the edges.

What kind of contams are we talking about? Meshed in mold? Mold satellites? Bacteria sliding around the plate riding with the colony? Different contams have different vectors and different ways to clean up so knowing the general taxonomy of what your dealing with would be helpful.

Just made the pic featuring contams my avatar. This is roughly 3 to 4 weeks after innoc. It was clean before the last four or five days.

Okay so you have mold riding along with the colony. That can be a real bitch. I wouldn’t pin my hopes on that colony, get more spores on to agar. The way to do it is knock up at least 4 or 5 plates with spores. Then only choose the best of those, toss the rest.

That's sort of what I did. I knocked up 4 plates with spores initially... all from the same print.
Are you saying I should start over from scratch?
Thanks btw.

Yeah if you have meshed in mold it’s often not worth the effort to save the culture. I would only bother if it was a killer clone I was trying to save.

Cool cool. I have no idea what the print is from. I did use cloth medical tape instead of micropore on the plates. Is that a potential vector for contamination? Also, I know I shouldn't have, but I wasnt using a clean box when i was making the transfers. My apt is super clean and it was during lockdown and i couldn't get to the store to get supplies to build a clean box.

Does a tek exist which lists the different ways to clean up agar based on contamination type?

Not for the most part. But for bacteria I like an agar sandwich. Meshed in mold I would try a warm pour. For yeast I would drop the temp to the mid 60’s. Antibiotics can provide an edge as well.

What about the duopore cloth tape for the breather hole on the plates? Is that not a good substitution for micropore tape?
None of the stores around me have micropore.

I don't know about the tape but are you saying you're doing these transfers just in open air? If that's the case you have to get some kind of still air box. It's not cleanliness that you're looking for its just a small closed in area with zero air movement. Then if you don't let your hands hover above your work you'll be able to do sterile procedures.

I've had regular Petris mold on the edges from using cloth tape. I'd stick with paper MP tape.
