You want to take from the very outer edge of those colonies, which are all the circles you see.  So look and find what you think is the best nice cleanest looking one, and then cut a tiny piece from the edge of it. You can take several pieces from different circles and colonies. You're then going to take that tiny piece of edge you cut and put it on a whole new blank agar plate to let it grow out.

I like the video. If i knew where you lived I would steal your roll of parafilm. I have to order it by the meter :shake: Also we use the same ziploc baggies ::pipesmoke:

With a plate like that, its pretty overgrown already, but pick a bigger colony and take a tiny (rice grain sized) wedge and drop it to a fresh plate. If a plate looks like that, then contaminates that are present (you dont know at this stage) might be hidden anywhere under that myc. so a good batch of transfers is in order. Once you see those grown out with around 2-3cm radial growth, post some pics and do leading edge transfers of those. going more than 2-3 transfers is only truly necessary for cleaning up a suspect culture or isolating further. I usually end up doing at least 4 transfers before hitting grains.

Hahah thanks! I'm going to start using parafilm but I had messed up and not used anything to seal my dishes. I lost 5 of em, but these survived so I'm babying em. Yeah, I'm going to probably end up doing 4 transfers so I have a real solid strain. Do you not recommend me throw these in grain once the agar is fully Inocculated? I don't want to have any waste.

If you dont want any waste, dont waste grain by putting inoculate you arent 100% sure of and turning it into neon soup.
