Mycelium transfer from contam plate to clean process in SAB

If I cut the clean myc out of the contam plate and close the lid and open the clean plate for a max of 5 seconds would It not spread the contam from opening the first plate? Do I need to make my cut in 5 sec max?

i think you’ll be fine man. you wanna be speedy obviously but i don’t think it really matters that much, i doubt by just opening it you’re gonna send spores or whatever into the air. i mean, people transfer from contam’d plates all the time. i’ve done it too & never had issues. unless your plate is literally all trich or something

There are spots of trich on the plate towards the outside but also clean myc in the center. I even thought of trying to use my oven as a flowhood because frankly The SAB isn’t that fun. In the market for a flowhood/FFU now

Well the purpose of the still-air box is to work with, as much as reasonably possible, still air
I don't think there is a time limit, in fact if you rush then you are probs more likely to create air currents w/ your movements. Bacterial colonies and molds cannot simply jump out of the plate. They need air currents to carry them out
As long as you work carefully and diligently in your SAB in a room without a running fan or close to a vent you will be fine

Gotcha, I want to say RR recommends no more than 5 seconds of open time with plates.. maybe for when using a flowhood ?

Yeah, I believe that's a general "rule of thumb" sort of thing. You definitely don't want to be dilly-dallying with your plates open any longer than you have to
But "have to" is the key phrase. If you're a newbie (like me) then you have not yet committed the movements (cutting wedges, streaking, etc.) to muscle memory so you cannot move as swiftly as an experienced hand
5 seconds is probably more than enough time for most people to cut a tiny sliver of myc from a plate, but not everyone is so coordinated. Me, for example, I've always had naturally shaky hands. Nothing I can do about it. Working with rice-sized pieces of agar will never be my strong-suit, so I compensate by making sure my work area is extra clean before I begin the steps that require swiftness
But yes, just find the right balance between speed and precision would be my advice. Good luck

i fuck up a lot putting wedges down off my scalpel on to new plates & i rarely ever get contams. i’m sure it’s not great to have your plates open for too long but even like 30 seconds is fine. idk if this helps but i tilt my plates to the side so they aren’t facing straight up. also hovering your hands over open jars/plates/upside down lids is what’s gonna be giving you contams more often then not i think
