My first grow after 6 years.
    #26811621 - 07/08/20 07:43 PM (3 years, 6 months ago)

Hi there, I started experimenting with mycology in 2014 but did not have much success. About 85% of my jars always got contaminated, and looking back now I threw a lot of jars out cause I though I was growing cobweb mold when all along it was tomentose mycelium. I have never had a successful monotub grow before so I guess that's what this thread is going to document.

I bought a ton of MS syringes and knocked up about 24 pf cakes about 3 weeks ago. My result was only 4 jars coming close to being fully colonized, and I know for a fact my sterilization/sanitation procedure is on point. I am use H2O2, Iso, bleach spray, 90 minute PC, and a SAB. So I decided its a good idea to start some agar work with one of my stalled colonizing pf cakes.

Took my first samples on July 7th

You already have bacteria, yeast and mold colonies on those plates.

are you sure thats not just vermiculite from the pf cake youre seeing? I took my samples right from my pf cake and put them in h2o2 to kill any bacteria. Shouldn't have that killed any bacteria?

I can upload a close up if you'd like.

Red looks like yeast or bacteria
Green is a potential mold satellite colony
I’m not sure if yellow is anything, maybe condensation and a less than clear pic.

The red and yellow is all condensation, I dont keep my plates flipped upside down, and that green circle you drew is actually a small crack in the petri dish, I dont know why it looks that way in the picture

The red is definitely not condensation unless it’s only milky in the pic. Still doesn’t mean it’s all toast. Might be able to clean something up. That’s what agar is for.

h202 is of questionable value.
i would clean/ upgrade the lab and pay more attention to procedure.
it appears to me you have a reasonably high ambient contam load in your work zone.
