Healthy looking and smelling spawns gives contam each time
    #26800836 - 07/03/20 06:51 AM (3 years, 7 months ago)

I have a PC that reaches 14.5 PSI so I sterilize my rye spawn for a bit longer than 2 hours.
I use a SAB and I try to follow all the cleaning sterile practices that I've read about.
With one exception, I no longer flame sterilize my scalpel, instead I put a scalpel and a small spoon which I use to inoculate with in a glass of bleach in my SAB. This way I don't need to take my hands out from the SAB and the tools are more evenly sterilized and stays in the SAB till I'm finished. This has helped me to reduce my trich rates a lot. I also don't use agar plates, instead I use sterilizable containers and I only open them once in my SAB to inoculate.

My agars usually turn out fine. Most of the time they are all evenly and tightly white but sometimes (and I don't know why) they are very pale or has almost nothing around the inoculating agar piece but otherwise white and smelling all sweet and shroomy.
My rye spawns sometimes look bacterial at the end of the colonization place and I fruit them (without luck) anyway because I've read people have had success with a bit bacterial spawns. Other times they look perfectly fine and also smell sweet and shroomy.
But I almost never have success to even get a decent first flush before I get contamination.

How can I tell whether or not a spawn is okay to fruit when it looks and smell fine?
And how can I eliminate this problem in the first place?

It shouldnt smell "sweet"

It should smell "earthy" and like mushrooms.

Bleach is not sterilizing your blade. Bleach disinfects it doesnt sterilize. I reccomend going back to using a flame.

When it looks really good the smell is a bit sweet, healthy sweet, not like contamination. Sometimes it's less sweet and more earthy but those times the spawn actually look worse.

Shouldn't it be sufficient to disinfect the scalpel then? I've tried reusing agar plates by cleaning them with bleach with success.
I will try to flame sterilize it before putting it in the glass of bleach this time to see if it's better. But I really want to keep this convienience of nothing having to go out from the SAB which has greatly reduced my trich rates.

Disinfecting and sterilizing are different things.
Yeah it’s suitable to disinfect but your not trying to disinfect you need to sterilize.
Flame sterilization only. Don’t flame sterilize then wipe with alcohol or bleach or any thing like that.
Flame the blade then use it.

Yeah, flaming the blade then putting it in bleach is pointless. Flame only. You use the agar in a plate to cool the blade.
