Question about old spores * 1
    #26798050 - 07/01/20 11:01 PM (3 years, 6 months ago)

I have panaeolus cyanescens prints that are 2 1/2 year's old. Should I rehydrate the spores before I put them on agar? Do you think I'll get growth without rehydrating them? For the most part they've been in the refrigerator the whole time. Thanks

People have been talking about forced rehydration using a rubberband and a syringe, I dunno how that works.

Never really had any germination when prints go for more than a year in room temp. But it might also be because of our very arid climate. Your spores might very well be viable because you stored it in the fridge. Id say streak some on agar and see for yourself.

Thats interesting. Does the rubber band go around the syringe and place pressure on the spores/syringe? :strokebeard:

OP, i would hydrate your spores in a syringe if i were u.

As far as I understand it where I've seen people talk about it is that the rubber band holds the plunger down creating pressure in the syringe. Which would then force the water in the syringe into the spores.

Logically, it makes sense. I have no idea if it works better than just normal rehydration lol. It would be interesting to measure spore size before and after and compare regular hydration vs. that syringe hydration. That also may be an awful way to test it lol:shrug:

I've never kept my prints in the fridge

I guess we shall see man, I just put some Panaeolus Bisporus on agar this morning, the print is at least 3 1/2 years old.
I just put the on agar with a loop. I didn't hydrate

Could you cap your half full syringe and pull the plunger back, then just wedge something or tape it there , creating a negative? Would that be the same as the rubber band wrapping?

No it would be the opposite

You want the contents to be under higher than normal pressure.
