very basic agar question

using a multispore syringe, whats the optimum number of plates to sample? as many as you can? or is there a point of diminishing returns? how many plates do you guys do?

As many as you can drip a single or close to it as possible drop on. No set number, just however many must be a few hundred. You don't want to use more than a drop if you can avoid it.

edit wait maybe I misread that. Do you mean how many transfers deep can you go or how many plates you can inoculate with a single syringe?

Oh I see what you are saying like how many would be a good number to start to work with a good gene pool? I do about 12 MS plates to start a new thang personally. And then I only pick one to transfer from and work on isolating out of that and leave the rest to pin invitro for back up clones.

I start 1 or 2 at a time when I germinate spores because if not I end up with a bunch of plates I don’t need.

Syringes can be tricky if you get ones like I’ve tried to use so maybe starting a few more would be beneficial. For prints though I use 1 or 2. Then you can transfer little bits of myc to however many new plates you want and it’s clean. With germ plates it’s hard to tell if they’re clean so if you have 8 successful plates and you transfer 2-3 times from a couple you still have all these crazy spore loaded fuzzy plates just hanging out.

thanks for the feed back.I heard this too. for what ever reason I get alot of weird looking tomentose cultures so far with mss to agar, and the ocassional trich. looking forward to when I can just be like

Im more or less starting a compost pile in the yard, older experiments and extra stuff go out there. including scraped plates, if you love it, set it free...
