limit to clone -> grain -> fruit -> clone cycle?
    #26794081 - 06/30/20 09:24 AM (3 years, 8 months ago)

I've read about limits to strain expansion via grain to grain, with problems creeping in if taken too far. I'm not likely to run into that, but what about the cycle of taking a clone to grain, then to bulk sub, to fruiting, then cloning to agar, transferring a few times, then back to grain and so on?

Is this likely to end up causing problems? And if so, I'm interested in understanding why... presumably limiting genetics too much? Assuming it is a problem, would taking prints and starting from spores every now and then break the cycle sufficiently?

If it matters, this is in the context of the GT variety, which I presume has pretty stable and limited genetics already.

That will not cause problems, just work clean. Every once in a while you should restart with spores but it would take months at the least before there was any noticeable affect.

Eventually, senescence will cause it to die (and presumably slow down/act different near the end). The problem is not limiting genetics, it's that nearly all complex organisms are programmed to die after enough cell divisions.

Senescence is not going to set in for a long long while, it's really not something you need to worry about.

I agree, I was just answering his question about why the clone -> grain -> fruit -> clone cycle cannot go on indefinitely.

I’ve read a couple times you shouldn’t keep making clones of a clone. Don’t know why, it’s just a common shroomeryism that might be untrue.

Doing it that way is unnecessary too if you just take a clone and then save the culture. You can keep a master slant or a thick poured plate in the fridge for a good long while. Whenever you need more you cut a wedge from that original (or the slave would be the safer way) and expand that out however you normally do.

That's a good point, just keep your culture on agar

Thanks for the explanations. Guess it will take a while if people can successfully expand 100x on grain without big issues. Will probably take me a while to get that kind of multiplication across agar, single jars, and small substrates. I'll take some spores in case a restart is ever warranted.
