Grow Log: Master Jar to G2G to Shoebox. Let the journey begin!

Grow Log:
I'm stubborn and tend not to follow directions well, but I always enjoy the path. Anyway, the intention of this grow log is to document the process I take in cultivating a single large WBS jar to allow multiple G2G transfers that will be utilized to cultivate 5 shoeboxes.
PHASE 1: Skip the soak and speed things up.
Well maybe "skip" is the wrong word, but I will be modifying the process and saving almost a whole day by cooking my WBS in a rice cooker to obtain decent grain hydration. - Sanitized myself with a few beers. - Strained out any floaters in the WBS (Might skip this step in the future seems messy and unnecessary. Will sunflowers and corn really destroy the world? I mean Monsanto, yeah, but come on what's the purpose?) - 2 cups WBS 4 cups water (just like cooking regular old rice - keeping it simple. It takes about 50 minutes and there will be some WBS left over. So, if you prescribe to waste not, want not, then shorten up your measurements to avoid dumping any excess WBS.) - Once WBS is "cooked," but more like speed soaked, I put it into the single large jar and prepare it for sterilization.
PHASE 2: Steam sterilization as a style choice.
I don't particularly like PC's, so I'm choosing to do one big steam sterilization of the single large jar of WBS. Splitting this process up into fractional steams would probably be better, but I'm trying to keep things moving forward by eliminating any unnecessary waiting time (this may have to be changed depending on contamination). - WBS is loaded into the single large jar, topped with vermiculite, and with some cheesecloth underneath the lid as a substitute for polyfill. - Scrunched up aluminum foil all around the lids edge to prevent any moisture from infiltrating the jar during the long steam. - Placed the jar into a canning pot with an elevated metal platform on bottom and brought water to a boil (took about 10 minutes), turned down to a simmer, then started the 6 hour timer. (Add pre- boiled water as necessary). - Once the 6 hours was completed I immediately put the WBS in the refrigerator for 3 hours (Might've gotten away with 2 hours, but I fell asleep, and yes I know that refrigerators are the Mecca of contamination and that patience is the key to this hobby, but let's just see what happens. Anyway, ideally I will eventually get a container that I can sterilize to place the single WBS jar inside, then stick it into the refrigerator in its own sterilized environment to quickly cool it down).
PHASE 3: Inoculate and wait.
I used a spore syringe that I bought online that is about 2 years old and was unrefrigerated - I know, hate me. - Sanitized the environment with isopropyl alcohol. - Sprayed the inside of the SAB with isopropyl alcohol. - Once the WBS had finished cooling, I gloved up, heated the syringe, and inoculated the jar of WBS with the entire spore syringe (more spores could potentially increase speed of colonization), then I placed tape over the four inoculation holes. - Told the spores to have beautiful dreams and grow big and strong, then placed the jar in the cupboard.
This whole process took me about 10 hours compared to the normal 48 hours or more with all the additional waiting time. If done right this could probably be knocked down to 8 hours which means you could have an inoculated master jar of WBS for G2G transfer much quicker than previously expected.

Can you please post pics to properly document this misguided attempt for future reference? Iām not sure why you think this was faster. You can have properly prepped grain ready for inoculation in about 2.5 hours. Your attempt to save time is only going to end up wasting it... https://www.shroomery.org/forums/showflat.php/Number/8508523

@Gastronomicus Thanks for the good luck. It is appreciated!
@Psilanthropist I'll post pictures of the results, otherwise I wrote down everything that I did. This process allowed me to prepare my WBS faster than normal by skipping a long soak period and speeding up the overnight cooldown with the refrigerator. 2.5 hours seems wildly fast when accounting for soaking and cooldown, but I could be wrong. Thanks for the nice WBS PC Tek.

While you're waiting for your mold to grow you should read more teks and buy a pressure cooker.

@Gastronomicus Pftahahaha! Thanks for the suggestion. Again I'm stubborn and I like steam sterilization. It's worked for me in the past. The rice cooker used for the "speed soak" and the rapid cool down in the refrigerator are the two new variables in my process. Fractional steam sterilization is underrated when done properly (which I didn't do since I just did one long 6 hour steam). I have confidence that the WBS will colonize. Regardless, I'll share a progress photo once any signs of anything show up on the WBS.

Using the whole syringe likely made the jar too wet and will slow things down.
Also you can just do a 2:1 ratio wbs:water and go straight into the pressure cooker. I usually fill the quart jar up to the 400 line with wbs then add a half pint of water. Pc for 90 mins and youāre done.
Iām all for experimenting and trying new stuff but youāll most likely end up wasting a lot of time not learning the conventional way first. Once youāve got experience with the basics then youāll know enough to start finding ways to speed things up that arenāt just a total shot in the dark.

Why vermiculite in a grain jar? And idk the micron or whatever rating of cheesecloth but polyfil works and is uber cheap. Sfd are cheap too, especially if you cut 7 quarter shaped circles out of a $1.05 90mm sfd.
If you actually want to grow mushrooms quick then Iād start some spores on agar and get a clean culture, prep your grain with Foomanās method or the no prep AKA just laid out, PC grain and a qt jar of water to make a blenderless LI with your clean colonized agar puck, pour that into your grain that will then be ready to spawn in 10 days +- Then make shoe boxes.
But if you want to do something just to make people go and see if you get anything... Iām in

Trying to stop a mold spore with cheesecloth would be like trying to stop a gnat with a chain link fence.

@A.k.a Very good point about the additional liquid from the spore syringe making the grain too wet. I didn't consider that as a possible issue. I'm staying away from PC and sticking with old fashion steam. Thanks for the input!
@Roger Clemency I was under the impression that a verm layer provides extra protection from bacteria. Is that wrong? In regard to the cheesecloth, I layer it up so that it is thick. I'm not putting a single thin sheet of cheesecloth or it would be exactly as Pastywhyte said about gnats and a fence.
@Pastywhyte Pftahaha! Great analogy.
Update: - No mycelium and no mold. - I will probably prepare a backup master jar, but approach the process with more patient since I'm down to my last spore syringe. I'll do a full rinse, soak (no rice cooker), steam, and a proper overnight cooldown, then inoculate. This way if the first master jar fails to colonize I'll be able to smoothly continue the grow log .

Update:
- The master grain jar that was prepared with the technique that I outlined above is showing signs of mycelium growth.

- As previously mentioned I decided to prepare a backup jar just in case things went south with my speedy preparation. The backup jar doesn't have a vermiculite layer on top as it was mentioned by Roger Clemency that it was unnecessary. I also prepared it in my normal manner that takes much more time which I will outline below (hopefully by seeing this preparation method you will understand why I sought to quicken the process).
Phase 1: The 12-hour soak.
There shouldn't be much more that needs to be said, but I'll notate everything as best I can.
- Strained/poured out all the sunflowers, corn, and floaters. - Rinsed the WBS until the water was fairly clear (I didn't even bother with this in the original preparation of the speedily prepared master jar). - Once the WBS was bathing in a clear bath of the finest tap water, I simply let it soak for 12 hours (note that this process was completed in 50 minutes with the rice cooker). - Finally when the 12-hour bathhouse treatment was completed I drained the WBS for about 30-45 minutes.
Phase 2: The 6-hour steam.
This process was basically the same as my previously speedily prepared master jar, but I didn't add a vermiculite layer nor put it in the refrigerator after the 6-hour steam sterilization.
- After WBS had drained, I loaded the jar with WBS, added layered up cheesecloth, put the lid on, then scrunched up aluminum foil all around the edge of the lid. - Brought water in my canning pot to a boil, turned it down to a simmer, then placed the jar into the pot and started the 6-hour timer. - Once the 6-hour steam was completed, then I turned off the burner, removed the pot from the heat, and let the master jar of WBS cool inside the canning pot for about 12 hours.
Phase 3: inoculate and wait.
I used a spore syringe from the same 2 year old batch that I used in the previous master jar, but I only put half of the syringe into this jar since A.k.a mentioned that the introduction of the additional liquid from the spore syringe may make the WBS too wet and slow colonization down. - Sanitized the environment with isopropyl alcohol. - Sprayed the inside of the SAB with isopropyl alcohol. - Once the WBS had finished cooling, I gloved up, heated the syringe, and inoculated the jar of WBS with half of a spore syringe then I placed tape over the four inoculation holes.
This preparation took me about 31 hours compared to the 10 hours it took when I utilized the rice cooker and refrigerator. Below is a picture of the backup master jar of WBS which again has no vermiculite layer and only half a syringes worth of spores.

Fyi, the width between cheese cloth fibers is about 500 microns. A bacteria is about 1-10 microns wide. Micropore tape has fibers less than 1 micron apart. I don't think the cheese cloth is going to hurt you here since you've (hopefully) closed the jar lid so tight that there's no air being exchanged through the cloth. But if you try to use cheese cloth for ge it will fail 100% of the time. Luckily you're using tape for ge

The verm layer is why the grow succeeded. Giving credit to the cheesecloth would be like putting a bandaid on a cast and crediting it with the bone healing.

@Pastywhyte Yeah that's quite possible. Since my backup master jar doesn't have a vermiculite layer we will be able to see the inadequacy of folded cheesecloth as a protective barrier/GE option. Hahahaa another quality analogy.

@metaphoric Below is the tape that I'm using and I don't believe that it is very breathable, but I could be wrong. I know in the past when I used scotch tape and no cheesecloth the mycelium didn't get enough GE and suffocated unless I removed the tape.
Anyway, after I inject the spores there is a large hole in the cheesecloth and that is why I would put scotch tape over the vulnerable hole, but there has always been a vermiculite layer in the cakes that I have made, so a good test would be to have a jar inoculated with a folded layer of cheesecloth, no vermiculite and scotch tape over the injection point. This would force the cheesecloth to be tested for GE and protection.

Test it however you want, cheese cloth won't work. Mold spores float freely through it. Cheese cloth is meant to filter solids, not microscopic particles.
Micropore tape is $3 at any pharmacy. Polyfil can be find in most pillows. I don't get why you insist on using nonsensical tools when proper tools are dirt cheap and available everywhere. But you do you, I'm out

PHASE 4: Grain to grain transfer to maximize mycelium.
Now that the single large jar of WBS is fully colonized it's time for the G2G transfer. - Prepared multiple smaller jars with WBS the exact same way as Phase 1-2. - Sanitized the environment and SAB the same way as Phase 3. - Inside the SAB I had all the new WBS jars and the large jar of colonized WBS. - I scooped some colonized grain into each new jar, capped them, then applied tape to the single hole on the smaller jars. - After each new jar had received colonized grain I kissed them and put them away in the cupboard.
Update:
- The first master jar that was prepared with a rice cooker and speed cooled in the fridge has successfully colonized. I used it to do 11 G2G transfers inside a SAB and I will post the results once they are fully colonized.
- The backup master jar that was prepared without the use of a rice cooker, cooled in the canning pot overnight and had no vermiculite as a filtration layer is almost fully colonized except the very top of the cake underneath the lid.

You know tyvek envelopes are free at the post office? And work as a sfd?
I'm curious about the thought process behind the grain and dry verm... sounds like you're combining a pftek and wbs tek...
I think everyone fills their wbs 1/2-3/4 of the way to leave room for shaking once its 30%+ colonized. Packing it full kind of defeats the purpose imo, why not just make brf cakes at that point?
Not to mention the dry verm is a contam barrier... that you have to go through for any gtg transfers...

Why not just put wbs in boiled water for 30 minutes then pc for 90 minutes after straining and drying? Alot faster than 6 hour steam sterilize. Your whole point here is to save time when 24 soak is old information nowadays. Anyways good luck

I feel like you answered for me.. inferior but half-ass functional is indeed better than useless.
I wasn't praising tyvek or saying everyone should use it, but the dude refuses to get a pc.. I figured he's trying to go cheap as possible..
Free is pretty cheap.
I also replied before reading some of your posts.. going to try non mod lids.. and you're a TC, and I'm a scrub... there's no doubt you know more.

@Cegda Yes, I'm aware that tyvek is a free option. Thanks for the recommendation.
I'm just kinda doing it my own way, but yeah, I did take the vermiculite idea from the pftek and apply it to WBS just as an extra precaution to minimize the likelihood of contamination.
As far as shaking the grain goes, well, I've never done it and everything has colonized just fine with previous grows. (I am aware that shaking apparently speeds up colonization).
Anyway, here is a close up picture of the backup WBS jar that I inoculated without the vermiculite layer:

Definitely has stagnated growth on the top possibly due to the failure of the cheesecloth to provide an effective barrier. Though it doesn't look contaminated. The mycelium seems healthy. I'll open it up soon and give it the good old smell test.

I was really wondering how GTG worked when you have to go through that dry verm layer. Its a contaminate barrier yes, but what are you doing before GTG? sticking a spoon through it sounds like a recipe for mold.

I was commenting on how you started negative and are still negative.. not trying to beef with you, and it seems that most people (according to your ratings) view you as very helpful.
I mentioned tyvek and you came at me.
I explained myself and even said I wanted to try your tek. You ignored that and were critical to op without providing any useful info.
And now I'm arguing with a moderator and TC and idk why. I'd much rather be reading your post and/or picking your brain.

I used to do a small square of tyvek held in place with micropore. That is much better than either one or the other as far as grain lid filters goes.

checkmatsis said: Grow Log:
The title of the post says master jar, which I associate with people who've had a long string of success, but I'm not getting that impression.
If I were you I would listen to this guy.

I dont bother to soak grains anymore. Seems like a waste of time and energy to me.
I use a 25 gal brew kettle to bring 50 lb of rye to a boil. Boil for 5-6 minutes then drain the kettle. The kettle takes about another 10 minutes to drain. Then scoop the grain from the kettle to the strainer. Once dry enough... bag it up.

@Pastywhyte It might not be as strong as rhizomorphic mycilium (colonizes faster and gives larger flushes), but I consider tomentose mycelium to be healthy. So, yes, different standards.
@Cegda I dump the dry vermiculite layer which exposes a mixture of mycelium and vermiculite, scrape off any excess, then I start digging into the mycelium from that region for the G2G transfer. I plop a scoop of the mycelium into the sterilized WBS and cap it. It's my understanding that a fully colonized substrate is fairly contamination resistant, so by keeping the colonized grain on top of the transfer jar it should help minimize contamination as long as the sterilization process was successful.
@LotKid Yeah, it might be missing some spokes, but it seems to roll lol
@JoBloShow I like steam sterilization. I use a rice cooker to get my WBS hydrated and it seems to work good enough and it avoids a 24 hour soak.
@ Brian Jones Yeah, I guess master jar might not be the correct term. What I mean by master jar is the originating jar that I colonized to use for the G2G transfer. Pftahahah, I own a pressure cooker, but I don't use it for anything. Anyway, there is nothing wrong with steam sterilization, experimenting, and enjoying the process. I'm simply sharing my experience.
Here is a progress picture of the 11 G2G transfer jars:

All of them seem to be colonizing beautifully.

I have zero issue with tomentose mycelium, most people who have grown RW will attest that itās pretty tomentose as a typical trait of the variety. Why would I have done that if I thought tomentose mycelium was unhealthy pretty sure I can tell the difference between tomentose mycelium and a reaction to bacteria.

@Pastywhyte Ah, yeah, I misunderstood your comment. I'm giving that jar some more time to colonize the top portion. If nothing changes by mid-week, then I'll do the smell test. Worst come to worst I'll remove the stalled section of the cake and use the healthy portion in the bulk grow.
@smalltalk_canceled I haven't gotten to fruiting yet. I'm sharing the process as I go along. So, I'm currently on the G2G transfer waiting for the 11 jars to fully colonize. Once that is finished, then I will be doing a shoebox inspired by ShaperDreaming SFF Shoebox Tek.

Wait, what? You own a PC, but you're choosing to steam sterilize grain jars instead?
What is your reasoning behind this? Are you trying to prove something?
That's not being stubborn. That's just choosing a sterilization process that is known to be less effective. You would never walk into a real bio lab and see steam sterilization being used. It will always be an autoclave. There's no need to debate this; it has been settled for years. But I'm just scratching my head trying to understand your reasoning here

@TheDoobsker It's simply how I'm choosing to do my grow and it has been working perfectly. Everything I have steam sterilized has colonized, so I'm completely happy with the results. There is nothing wrong with steam sterilization. If it didn't work, then it wouldn't be an option. Besides, how do you skin a cat? Anyway, I'm not debating anything. I'm simply sharing my current grow.
@Cegda As far as using a pressure cooker to save time, yeah, that is fully true. I might go rice cooker, pressure cooker, then refrigerator to see how fast I can prepare WBS for inoculation on my next grow.
@LotKid Pftahahaha very true.
I'll post a picture of the results of the G2G transfer once they have finished colonizing.

Man, u cant be serious. Stop trying to defend this this verbally, and wait for the pressure cooker in silence. Then use the pressure cooker.
Refusing the PC is one of the most scrub things in this hobby, woefully ignorant, provocative to the established, and inferior as method both in power and flexibility. Dont even reply. Just call the responsible parties, and ask: "When is my pressure cooker coming?"

How many jars fit in your rice cooker vs the pressure cooker? Look at no soak teks if that's your problem with seeds. You will have sterilized seeds in alot less than 6 hours. But hey I'm glad you are having success so far even if its unconventional

@smalltalk_canceled Yeah, uh ok, I'm not defending anything. Don't get bent out of shape. I'm simply sharing my current grow. It could all go horribly wrong, then everyone can celebrate, but so far it has been successful.
@TheGreatGig No jars go into the rice cooker. I simply cook my WBS in the rice cooker to get them hydrated before sterilization and inoculation. My HUGE canning pot can hold 11-14 jars (more or less depending on the jar size). I can leave the jars steaming for 6+ hours without adding water. I'll have to check out some no soak teks. Thank you, yeah, I should have made the thread name: Unconventional Grow Log (Trigger Warning!) hahaha!

Update:
The 11 G2G transfer jars are just about fully colonized without a single contamination. In about a week, to allow the mycelium to consolidate, I will prepare coconut coir for some shoeboxes. Everything is moving right along perfectly.
To clarify: I didn't shake these G2G jars and kept the transferred mycelium on top of the steam sterilized grain and let it colonize downward.

Phase 5: SHOEBOX TIME
Come along with me and the butterflies and bees, we will make some shoeboxes and grow some mushies.
Anyway, I more-or-less followed SFF Coir Tek to prepare my bulk substrate, then squeezed the living hell out of the coir to get it to "just under field capacity," mixed some colonized WBS with some coir, then added the "pseudo-casing" of coir on top of the mixed mess. Once that was completed I sprayed about 60 sprays of distilled water on the coir to create the luxurious top of the box steam room just as it is discussed here: SFF Shoebox Assembly TEK. Anyway, so far this entire process has been successful, but there are things that I will change on my next grow. Finally, no matter what the results I will post the outcome.

Side note: I saved one large jar of WBS and one smaller jar to continue the process starting with some G2G transfers just incase everything goes south.

Good luck man! Hope it works out. I just harvested ~80 wet grams from a grain shoebox.. the potential is there!
I've been using spitballs cvg tek. Just follow the recipe and no worries about field capacity. 3qts of spawn works out to 200 grams coir, 2.5 cups verm, 73 grams gypsum, and 1300 ml water... I've had nice results with 1000 ml water and 300 ml coffee as well.
Just mix in a bucket with a lid and let it sit for a few hours before making the box. 3 quarts is about 1 shoebox for me.

Those are pretty crazy ratios, thatās like 10 quarts of spawn to a brick of coir.

Definitely curious about your yield per shoebox. Your boxes look nice!
I made a shoebox on august 15th using the same tek. I may have over-misted but it came out to about 300grams wet for the 1st flush. 2nd flush was a lot less though @160g wet.
I also just spawned another one and did 60 sprays as you did (a lot less than my first box). I'll update how it compares.
Shoeboxes are great

Iāve never argued this...I only suggest that the precise methods produce more mushrooms than the hamfisted ones. Anyone can grow a few weeds, itās the canopies that separate good methods from poor.

Phase 6: Harvest, rehydrate, conclusion and continuation.
- The total harvest was 750 wet grams. 150 less than I anticipated. I may have harvested too early.

- Rehydrated for about 4 hours.
- Though I used folded cheesecloth as a substitute for polyfill it was definitely the 1-inch layer of vermiculite that provided protection from contamination. The backup master/starter jar that I prepared with only cheesecloth as a protective barrier colonized, but had bacterial issues and once put to a bulk substrate succumbed to trichoderma.
- In the future, I will continue to use vermiculite on my WBS, but I will couple it with micropore tape at the four injection holes. Two is one, one is none.
- The 11 jars that I G2G transferred were all successful and I believe that was due to keeping the colonized grain on top of the sterilized WBS and not shaking the half-pint jar at all. This is because mycelium itself is resistant to contamination when fully colonized. This allowed the mycelium to protect the WBS below as it grew down into the half-pint jar.
- My next steps will be to do more G2G transfers and shoeboxes. I will hydrate the WBS in a rice cooker, then load into jars, and steam sterilize the WBS for 6 hours (though for speed's sake a PC would be more beneficial). These jars will not have vermiculite on them and will be allowed to cool overnight in my giant canning pot, then I will move them to my SAB and do the G2G transfers.
- Any grows starting from scratch will be done with a starter jar (apparently master jar is the incorrect term) that has a 1-inch layer of vermiculite and micropore tape covering the injection holes. Start by rice cooking the WBS to hydrate, load into the starter jar, then steam sterilize for 6 hours (recommend PC for speed), then I will place the jar in the refrigerator for a speed cool down for about 3 hours. Once it has been cooled I will then inoculate with a spore syringe inside a SAB. Once this jar is fully colonized, then I will proceed to G2G transfer and shoeboxes.
- Because I am making my own spore syringes it would be best to screen the spores on agar to minimize contamination, wasted time, and to optimize genetics. I could be wrong, but it would make sense that isolated genetics would be a contributing factor in producing better yields and the coveted canopy.
Last photo of the best shoebox before harvesting:

Also, going to try switching out the shoebox cover for a plastic bag once pins start showing on this next flush to allow for a potentially taller grow.

I'm really only doing it to give myself a point of reference so I can reduce or increase the amount I spray it in subsequent grows as needed. Once I feel I'm getting the moisture right so I can set & forget, I'll stop counting.
The tek says to spray for 20 seconds but due to variances in time between sprays, spray bottle types, and human error & different climates, I figure it is best to figure out how many sprays works for me and then work off that from memory.
It's not a permanent thing haha - just using heuristics to help me learn until I don't need heuristics

@Rhizomorph Exactly, 20 seconds is variable, hence the counting and eventual dialing it in by eye. Experience is what gives the finger tip feel, or the ability to recognize contamination and optimal grow conditions.

The second flush was only 434 wet grams.

On this flush, I did switch out the shoebox lids for grocery bags and it seems to allow the fruits to grow a bit taller and potentially thicker (perhaps due to the increase FAE).
For the third flush, I decided to spray 60 sprays of water directly onto the colonized substrate, drained out any excess water, and I will probably swap out the lids again for plastic bags once I notice pins.

I've noticed my fruits tend to be somewhat thin from the SFF shoebox tek with the lid latched on, but this may be genetics as well since I'm running MS until I isolate some stronger cultures. Gonna try switching to a plastic bag once pins show up on the box I'm running now.
How big/how many holes do you put in the bag? In Bod's shoebox tek he doesn't specify and I haven't tested out what works best for me yet.

@Rhizomorph Yeah, I've had similar results using MSS of Golden Teacher with shoeboxes. You have to be sure to unlatch the top when the fruits are near popping, but even with doing that there is resistance from the lid that prevents the mushrooms from growing taller, hence switching out the lid for a plastic bag. I got the idea from Bod's Shoebox Tek *thumbs up*.
I used target bags. It was big enough to stretch/wrap around the shoebox fairly tightly. If possible, use a clear/see-through bag as it will be easier to see the mushrooms grow and when you will need to harvest. The target bag was dark grey and I couldn't see anything lol.
Two of the bags had thumbnail size holes in the tippy top and one of them seemed to not have any holes. I don't believe these bags are airtight, so there is not much need for modification, but the ones with the thumbnail size hole seemed to do better.
Please share your results!
@smalltalk_canceled Yes, during this grow I aimed to make 5 shoeboxes, but only made 3. So, I am combining the weight of 3 shoeboxes when I report the wet weight. The current total wet weight from the 3 shoeboxes is: 1,184 grams.
