Grow Log: Master Jar to G2G to Shoebox. Let the journey begin!
    #26777190 - 06/23/20 04:10 PM (3 years, 7 months ago)

Grow Log:

I'm stubborn and tend not to follow directions well, but I always enjoy the path.
Anyway, the intention of this grow log is to document the process I take in cultivating a single large WBS jar to allow multiple G2G transfers that will be utilized to cultivate 5 shoeboxes.

PHASE 1: Skip the soak and speed things up.

Well maybe "skip" is the wrong word, but I will be modifying the process and saving almost a whole day by cooking my WBS in a rice cooker to obtain decent grain hydration.
- Sanitized myself with a few beers.
- Strained out any floaters in the WBS (Might skip this step in the
future seems messy and unnecessary. Will sunflowers and corn really
destroy the world? I mean Monsanto, yeah, but come on what's the
        purpose?)
- 2 cups WBS 4 cups water (just like cooking regular old rice -
keeping it simple. It takes about 50 minutes and there will be some
WBS left over. So, if you prescribe to waste not, want not, then
shorten up your measurements to avoid dumping any excess WBS.)
- Once WBS is "cooked," but more like speed soaked,  I put it into
the single large jar and prepare it for sterilization.

PHASE 2: Steam sterilization as a style choice.

I don't particularly like PC's, so I'm choosing to do one big steam sterilization of the single large jar of WBS. Splitting this process up into fractional steams would probably be better, but I'm trying to keep things moving forward by eliminating any unnecessary waiting time (this may have to be changed depending on contamination).
- WBS is loaded into the single large jar, topped with vermiculite,
and with some cheesecloth underneath the lid as a substitute for
        polyfill.
- Scrunched up aluminum foil all around the lids edge to prevent any
moisture from infiltrating the jar during the long steam.
- Placed the jar into a canning pot with an elevated metal platform
on bottom and brought water to a boil (took about 10 minutes),
turned down to a simmer, then started the 6 hour timer. (Add pre-
          boiled water as necessary).
- Once the 6 hours was completed I immediately put the WBS in the
refrigerator for 3 hours (Might've gotten away with 2 hours, but I
fell asleep, and yes I know that refrigerators are the Mecca of
contamination and that patience is the key to this hobby, but let's
just see what happens. Anyway, ideally I will eventually get a
container that I can sterilize to place the single WBS jar
inside, then stick it into the refrigerator in its own sterilized
environment to quickly cool it down).

PHASE 3: Inoculate and wait.

I used a spore syringe that I bought online that is about 2 years old and was unrefrigerated - I know, hate me.
- Sanitized the environment with isopropyl alcohol.
- Sprayed the inside of the SAB with isopropyl alcohol.
- Once the WBS had finished cooling, I gloved up, heated the syringe,
and inoculated the jar of WBS with the entire spore syringe
(more spores could potentially increase speed of
colonization), then I placed tape over the four inoculation holes.
- Told the spores to have beautiful dreams and grow big and strong,
then placed the jar in the cupboard.

This whole process took me about 10 hours compared to the normal 48 hours or more with all the additional waiting time. If done right this could probably be knocked down to 8 hours which means you could have an inoculated master jar of WBS for G2G transfer much quicker than previously expected.

Can you please post pics to properly document this misguided attempt for future reference? I’m not sure why you think this was faster. You can have properly prepped grain ready for inoculation in about 2.5 hours. Your attempt to save time is only going to end up wasting it...

@Gastronomicus Thanks for the good luck. It is appreciated!

@Psilanthropist I'll post pictures of the results, otherwise I wrote down everything that I did. This process allowed me to prepare my WBS faster than normal by skipping a long soak period and speeding up the overnight cooldown with the refrigerator. 2.5 hours seems wildly fast when accounting for soaking and cooldown, but I could be wrong. Thanks for the nice WBS PC Tek.

While you're waiting for your mold to grow you should read more teks and buy a pressure cooker.

@Gastronomicus Pftahahaha! Thanks for the suggestion. Again I'm stubborn and I like steam sterilization. It's worked for me in the past. The rice cooker used for the "speed soak" and the rapid cool down in the refrigerator are the two new variables in my process. Fractional steam sterilization is underrated when done properly (which I didn't do since I just did one long 6 hour steam). I have confidence that the WBS will colonize. Regardless, I'll share a progress photo once any signs of anything show up on the WBS.

Using the whole syringe likely made the jar too wet and will slow things down.

Also you can just do a 2:1 ratio wbs:water and go straight into the pressure cooker. I usually fill the quart jar up to the 400 line with wbs then add a half pint of water. Pc for 90 mins and you’re done.

I’m all for experimenting and trying new stuff but you’ll most likely end up wasting a lot of time not learning the conventional way first. Once you’ve got experience with the basics then you’ll know enough to start finding ways to speed things up that aren’t just a total shot in the dark.

Why vermiculite in a grain jar? And idk the micron or whatever rating of cheesecloth but polyfil works and is uber cheap. Sfd are cheap too, especially if you cut 7 quarter shaped circles out of a $1.05 90mm sfd.

If you actually want to grow mushrooms quick then I’d start some spores on agar and get a clean culture, prep your grain with Fooman’s method or the no prep AKA just laid out, PC grain and a qt jar of water to make a blenderless LI with your clean colonized agar puck, pour that into your grain that will then be ready to spawn in 10 days +- Then make shoe boxes.

But if you want to do something just to make people go :horrified: and see if you get anything... I’m in

Trying to stop a mold spore with cheesecloth would be like trying to stop a gnat with a chain link fence.

@A.k.a Very good point about the additional liquid from the spore syringe making the grain too wet. I didn't consider that as a possible issue. I'm staying away from PC and sticking with old fashion steam. Thanks for the input!

@Roger Clemency I was under the impression that a verm layer provides extra protection from bacteria. Is that wrong? In regard to the cheesecloth, I layer it up so that it is thick. I'm not putting a single thin sheet of cheesecloth or it would be exactly as Pastywhyte said about gnats and a fence.

@Pastywhyte Pftahaha! Great analogy.

Update:
      - No mycelium and no mold.
      - I will probably prepare a backup master jar, but approach the
process with more patient since I'm down to my last spore syringe.
I'll do a full rinse, soak (no rice cooker), steam, and a proper
overnight cooldown, then inoculate. This way if the first master jar
fails to colonize I'll be able to smoothly continue the grow log .

Re: Grow Log: Master Jar to G2G to Shoebox. Let the journey begin! [Re: checkmatsis]
    #26789605 - 06/28/20 11:26 AM (3 years, 6 months ago)

Update:

- The master grain jar that was prepared with the technique that I outlined
above is showing signs of mycelium growth.

- As previously mentioned I decided to prepare a backup jar just in case things went south with my speedy preparation. The backup jar doesn't have a vermiculite layer on top as it was mentioned by Roger Clemency that it was unnecessary. I also prepared it in my normal manner that takes much more time which I will outline below (hopefully by seeing this preparation method you will understand why I sought to quicken the process).

Phase 1: The 12-hour soak.

There shouldn't be much more that needs to be said, but I'll notate everything as best I can.

- Strained/poured out all the sunflowers, corn, and floaters.
- Rinsed the WBS until the water was fairly clear (I didn't even bother with
this in the original preparation of the speedily prepared master jar).
- Once the WBS was bathing in a clear bath of the finest tap water, I simply
let it soak for 12 hours (note that this process was completed in 50
      minutes with the rice cooker).
- Finally when the 12-hour bathhouse treatment was completed I drained the
      WBS for about 30-45 minutes.

Phase 2: The 6-hour steam.

This process was basically the same as my previously speedily prepared master jar, but I didn't add a vermiculite layer nor put it in the refrigerator after the 6-hour steam sterilization.

- After WBS had drained, I loaded the jar with WBS, added layered up
cheesecloth, put the lid on, then scrunched up aluminum foil all around
      the edge of the lid.
- Brought water in my canning pot to a boil, turned it down to a simmer, then
placed the jar into the pot and started the 6-hour timer.
- Once the 6-hour steam was completed, then I turned off the burner, removed
the pot from the heat, and let the master jar of WBS cool inside the
canning pot for about 12 hours.

Phase 3: inoculate and wait.

I used a spore syringe from the same 2 year old batch that I used in the previous master jar, but I only put half of the syringe into this jar since A.k.a mentioned that the introduction of the additional liquid from the spore syringe may make the WBS too wet and slow colonization down.

- Sanitized the environment with isopropyl alcohol.
- Sprayed the inside of the SAB with isopropyl alcohol.
- Once the WBS had finished cooling, I gloved up, heated the syringe,
and inoculated the jar of WBS with half of a spore syringe
then I placed tape over the four inoculation holes.

This preparation took me about 31 hours compared to the 10 hours it took when I utilized the rice cooker and refrigerator. Below is a picture of the backup master jar of WBS which again has no vermiculite layer and only half a syringes worth of spores.

Fyi, the width between cheese cloth fibers is about 500 microns. A bacteria is about 1-10 microns wide. Micropore tape has fibers less than 1 micron apart. I don't think the cheese cloth is going to hurt you here since you've (hopefully) closed the jar lid so tight that there's no air being exchanged through the cloth. But if you try to use cheese cloth for ge it will fail 100% of the time. Luckily you're using tape for ge

@metaphoric Thanks for the information! I will probably switch to polyfill in the future, but this current method has been working for me. Last time I used a vermiculite layer, cheesecloth, and scotch tape and got beautiful colonization. Also, yes the lids are snugged down tightly.

The verm layer is why the grow succeeded. Giving credit to the cheesecloth would be like putting a bandaid on a cast and crediting it with the bone healing.

@Pastywhyte Yeah that's quite possible. Since my backup master jar doesn't have a vermiculite layer we will be able to see the inadequacy of folded cheesecloth as a protective barrier/GE option. Hahahaa another quality analogy.
