Sterilising Agar without PC?

I've been hunting for my first PC recently, and not had much luck (in part because the world is on hold and a lot of places are out of stock, add that to the fact that the UK sucks for PC's). When I finally found a suitable one (actually it was an autoclave), They emailed me a few days later to say it was out of stock!
Anyway, drama's aside, i'm seeking some insight as to whether its worth having a crack at either pasteurisation or tyndallization for sterilising agar? I've essentially had everything I need - all bar a PC - for a couple of weeks now and I just want to crack on at last! I hate the idea of having to wait god knows how long before I can get a PC.

Luckily liquids sterilize a lot faster than things like grains -- I have done Tyndallization on agar before, but without the extra day cycles, and had clean agar... Here's what I did:
- Take your bottle or whatever of agar that you intended to PC (I used a 1/2 pint mason jar, so the agar was close to the boiling water... sucks to pour though)
- Get a big pot (I used a 9" diameter pot) and some kind of trivet (higher is probably better so you can get more water in there). I had mine about 2 inches off the bottom of the pot.
- Fill the water up to about 1/2 an inch above the bottom of the jar or bottle or whatever
- boil (like a proper boil) for an hour. I had to refill the water half way through to keep the water at least touching the jar.
Worked for me a few times, can't guarantee that it will work for you, and obviously PC'ing is the proper way to do it

Great, that's good to know i'll try that out. Does it matter how much I fill them up? My thinking is not to fill them too high, so as to avoid potential overflow. The bottles i'll be using are the size of small stubby beer bottles.

The media bottles you mean? I wouldn't fill them up too high (but probably doesn't matter), and still wrap the top with foil to avoid condensation dripping into them.
Honestly I have no idea what is or isn't a good idea for this, I just tried this for about a month when I had no pressure cooker and it seemed to work for agar at least. You might have to experiment and see if the plates are clean in several days =/

Look on local sales sites in your area..second hand is better than nothing.You need a PC mate..it makes life so much better in this hobby.

Why not buy premade kimmig petri dishes in the meantime until you have the proper equipment. They go around 2$/dish around here. Make sure you don't buy them from some garage lab/shop, the proper ones are gamma irradiated to sterilize them and can last for many months.

Well last week I FINALLY managed to get hold of one! I'm yet to test it out, but its a 9L Futura PC, and only set me back £79

Well I bought some of borosilicate ones not long ago. As far as I can tell, now that I have a PC at last, I now have everything I need.
Wish me luck! You know i'll need it! Haha
