Rehydrating Spores - Emergency Revival after 8 years of Storage?
    #26750771 - 06/17/20 05:54 AM (3 years, 7 months ago)

1.have some cube spore prints that are ~8 years old and tried to
germinate them on agar without success...
its been a month and 0% growth occurred.

2.loaded a steam sterilized spore syringe with a water/spore combination and let it hydrate for 7 days.

injected into PF cake and agar.
no growth only white glibber which most likely is bacteria.

any advice on how to get the spore prints to germinate?

maybe LC with LME , then on agar?

What's your agar recipe? I find a softer agar helps sometimes for germinating spores. My normal is 20g agar per 1L of water. I havent done it in a while but if I'm remembering right I start off around 16-17 g agar per 1L, give or take for spores that wont germinate.

  1 liter  water
  20g agar

by using 3-4g of less agar you think it will work?

Try different Agar recipes, maybe the LC. Spores should be viable for years. I’ve seen people successfully germinate 17-year-old spores.

Taken from
TL's Simple Liquid Culture (LC) Tek (Recipe) Using Two Ingredients (ELME)

3g ELME per 600mL water
sterilize this inside a jar.
inject spore syringe into it.

let mycelium colonize the LC culture and transfer to agar?

Some people have had success forcing water into the spores by putting a rubber band on the plunger to increase the pressure inside the syringe.

tried that but didnt seem to work still no germination.

what about heating up the spores in water at a specific temperature for some time?
