Cloning, isolating, and crossing
    #26746452 - 06/15/20 12:13 PM (3 years, 7 months ago)

Have a list of questions that I'm confused on and would like a better understanding of it

1) How do you 'isolate genetics/certain traits'? From my understanding, after doing multiple transfers on agar and growing each and every one of them, you go back to the dish that yielded the traits you desire (ex, canopy, large, potent, etc). However, what confuses me is that even if you do back to that plate, there are still many sectoring and genetic diversity with the mycelium. So even if you go back to that plate and start a new grow, it doesn't ensure the same results as the initial grow.

2) Expanding on question 1, the solution to genetic diversity is probably monocultures. So do I skip the step of growing out each sector, and instead, do as much transfers as needed until I get monocultures and then grow out each monoculture?

3) If you grow out a cloned tissue, can you further isolate genetics by cloning the clone? Or do you have to "reset" and take a spore print and do spore to agar and grow out each sector and then clone favorable mushrooms, and repeat the process? Would that method get me closer to a monoculture with traits I've selected to clone throughout the process?

4) I get insanely huge mushrooms on my later flushes. Would cloning later flushes yield large mushrooms too?

5) How do I cross 2 different varieties and stabilize it? Let's say I want to cross B+ with PE, do I put spores, mycelium, or clone tissue from each variety and place them in 1 petri and hope they germinate?

Crossing can happen easily because they are the same species. Easiest method drop two jars of two different cultures with different visual differences in the same shoe box. Hope for the best. Then grow it out from the prints. Usually F1 reverts back to one of the parents but as you get down the line you will see a dominate pheno and then clone that and print from it and keep going. All the other stuff about monoculture is really hard to pull off with out a lab.

I’ve seen people drop spores of 2 different varieties onto agar (on each side of the plate), let the plate colonize to the middle and what’s supposed to happen is new growth should go down the middle, creating a cross. There’s much more than that but it’s the gist of it.
