Can you store a leftover spore syringe safely?

Hello, I really just have that as a question. I was hoping someone could tell me if its safe to do so. I have 8 wbs jars ready to be inoculated but also have agar that will be nocced up in the coming days when i get more iso alcohol. (cant manage to find any at all anywhere right now) Assume that I have to store them and cannot do agar along side the noccing up wbs qts.

Can I safely use 8ccs in a syringe and leave the needle attached but place the cap back on it and store it until I need to use again?

My concern is I will leave the syringe contaminated somehow.

P.S. Its my first attempt with agar after having so much trouble after 5 years of doing this. I cant seem to consistently succeed with simple MS syringe + wbs.

You cant get ethanol, everclear, or else bleach water is just as good imo, it just doesn't evaporate like alcohol.

Well, thanks for your input sincerely :smile: but I asked if I could safely store the syringes after inoculating some WBS quarts but to answer your question. I have bleach and yes I have water so I could do that but I've never researched what the benefits of using bleach water to clean for example, the jar lids before inoculating via spore syringe, vs using straight up iso alcohol to do that.

Im always 100% ready to change anything Im doing if its harmful to the outcome. Can you really just use 1 part bleach and 10 parts water mixture to the same effect as 70% iso alcohol?

You could cap the syringe but id be weary of leaving the needle on it capped, you have no barrier between the syringe and the outside. I think the concentration is 5 tablespoons bleach to 1gal h2o. It takes 7 min of contact to kill everything so spray it on let it sit for a few minutes then its good to go.

Hmmm, i will likely do the bleach mixture after researching that a bit more. I was thinking I could cap it after flaming the needle again and just leave it in a ziploc bag until needed which will only be a day or two at most. I would of course place them back in the fridge. But since I will be using the leftover for agar would it be more viable considering any contams that show up, I could transfer away from by (if any appears) using the healthy myc that shows up and transferring that to a new agar plate and so on.

Yep as long as the syringe is not at a high contamination level when you put it away no harm, but you should be able to clean it up on agar.

Seems pretty obvious that the bleach would do the better job. Was that diluted bleach though or straight up bleach with nothing mixed in?

You should be fine storing it with the same needle and cap, since you'll be flame sterilizing your needle before using it again the next time. However, if you plan on not using the syringe for quite a while, you may want to consider the fridge. Though I did hear somebody mention that the switching from room temp to cold, then back to room temp opens the spores up to possible contam. Not sure if that's correct information or not, cheers

bleach and alcohol Is not that effective straight with out water.

70% alcohol works better than 92% alc. (alcohol Is so hygroscopic you won't  see It above 92% unless you get a lab grade)

It's the water that promotes the deterioration of the microbe cell walls.

Store used syringe in the fridge after use with cap on it before you inoculate the jars wipe whole syringe down wit 70% iso flame sterlize rewipe wit alch to cool down an inoculate in a clean sab also whip your jars down before inoculation as well

Dont wipe your needle after you flame sterilize it, that defeats the purpose of flame sterilizing the needle. Put a drop or two of the contents inside of the syringe through the sterilized needle to cool it.

Ya I heard you can do that I figured it was just as effective. an for defeating the purpose I see where your coimg from. I just figured it was an extra safety barrier to flame sterlized. Then the iso to cool down quick as a safe way. I haven't worked wit them in awhile.that's just how I did it still use sometimes tho
Haven't had a problem from it yet anyways

You don't see dr's dipping a sterile utensil into any type of sanitizing agent before going to work on something that they don't want to deposit anything into. Once the utensil is sterilized you don't need anymore sanitize, and all that alcohol is going to do is evaporate off and potentially contaminate the sterile surface. It's just not a good idea to dip a sterile needle into a alcohol solution before you are ready to inoculate something. Storing tools in alcohol is not a bad idea in between transfers and what not, just not right after you sterilized it.

Ok ya that's just how I learned. but what your saying makes sense.i don't know if doctors flame sterlize tools before working.figured they came in a pre sterlized pack.sn I only wipe my scaples wits iso then flame sterlize then right to dish.only have done it with syringes.

No dr's open a sterile pack of tools and don't sterilize them again, they are already sterile. And I wouldn't want to be hacked open by a flame sterilized scalpel that would leave carbon all over inside of the wound. The same principles are still in play when it comes to sterile procedure, no matter if you are working on a petri dish or a person all the rules still apply.

Ya that makes sense. An they wipe the skin of the patients wit iso. Not the tool cause it's already sterlized.same as u wipe the qt jar so it's sanitized but why sanitize a sterlized tool.i get what your saying
