New to Liquid culture, Need suggestions

I have agar down, and I want to try liquid culture.
What are some good teks? I searched but most of the info was older.

At it's simplest you are just dropping a wedge into LC broth and letting it colonized.
Personally I use Light Malt Extract + water for my LC broth.

It's a good idea to buy an inexpensive magnetic stir plate. It keeps your LC from forming stringy blobs.
There is also no need for fancy lids since you won't have to break it up. I like to use plastic one piece lids and pop an SFD underneath or use poly fill, just a basic GE filter like for a spawn jar.

I like to blend my wedge with water in a sterilized Eberbach unit and then pour into my LC broth to sit on stir plate. It's typically fully colonized in 3-4 days.
Could also just blend wedge in water and use as a liquid inoculant.

You've done the hard part. From Agar to liquid culture is easy.

I'd recommend 1% light dry malt extract:

Magnetic stir plate helps but you also shake them manually, if you don't want to buy/build the stirrer.

My favorite thing about the magnetic stir plate is that you don't need any special lids, poly fill or SFD work fine because there is no reason to shake up the LC to break up the culture

You can do manual stirring without a special lid, if you fill your LC  jar only half full. I'm forced to do this now as I dumped all my stir bars in the toilet by mistake. Magnetic stirring is definitely better.

I really like your Eberbach unit. Can you just pressure cook it for sterilization?

Yeah man! That's what they're made for.
They are crazy expensive (3-500$ for the container alone) but if you keep searching eBay I see them up for peanuts occasionally when the seller doesn't know what they are worth. Replacement blades, gaskets and lids are really cheap if you need to get new ones for one.

I have a stainless steel 250ml unit too with a press lid but I think the glass unit is better for our purposes. You can really damage the myc if you blend too long and it helps to see. Plus it's a 500ml container and it's smoother opening And closing the cap. But both are great.
They fit any Waring blender base but if you can find the lab blender with the seconds timer it is great because you can be really consistent that way. 



I think the glass unit you have to loosen the lid when you sterilize it, the metal unit with the press fit lid you do not need to.

I am wondering if I should keep using honey lc which worked OK for me but haven't tried anything else yet. I have had a few glasses stalling but nothing crazy. The reason I do LC at all is to have faster colonization than agar to grain.
So are other types less prone to grow contams? Can I overcook grain water lc to be on the safe side since there is no real sugar in it?

You are inoculating your LC with agar collection yeah?

Are you fellas able to use LC without contaminating all your bags or jars? Its quite scary that you cannot see contams like you can with Agar.

Recently, I have been doing agar to grain, then Grain to Grain, and colonization times are so much faster its awesome. some bags are done in 2 weeks, rather than 3-4 weeks with Agar or syringes.  I was doing agar to grain for all bags previously.

Im finally loosing less bags, lower failure rate. I'm somewhat new and I was loosing 30% of my bags almost every run, after what I read about LC im kinda scared to use it honestly, but I still want to give it a shot.

Are the colonization times about the same for Liquid Culture or Grain to Grain?  I was using around 1/2qt grain per jar on transfers.

If you can do g2g you can do lc. I just make however much I need for the batch and pour it all in one session.

Using it then putting it back does make me a little nervous but I’ve never actually contaminated one from using it yet that I know of.

Lc can be even faster than g2g. I just had a jar colonize from lc in 5 days. Usually it’s 7-9 days and g2g depends how much you use but 10-14 for me usually.

So storing LC in the fridge is not good? is it something that has be used immediately?  Damn 5 days would be awesome!

No no fridge is fine.

I just like to use it all at once when I open it because you never know if a contam got in there and then you put it back and it’s no good later.

I have used some a few times over the course of months but I have so many varieties now I stopped doing that.

Now I drop a wedge into the broth and let it sit at least 5 days then use it all at once. I normally make 200-300ml.

It takes some figuring out like everything in cult but it’s awesome when you get it down. So much faster. No waiting on plates to grow, soon as you have a tiny bit of clean myc you can drop it.

I went from taking a wedge off a plate to spawning 6 jars in 10 days last time. The plate I used hadn’t even grown out enough to knock up six jars by the time I was spawning.

I am using 5qt bags, would they still possibly colonize in as little as 10 days using LC?

Myself I use 2ml or so for a quart. I often free pour.

For a 5qt bag it's possible. Depends on the culture's genetics, is it clean - is there any bacteria, how you prepped your grain, how much GE it gets, temp..

You get the picture. Few days for the LC to recover + colonize enough grain for a shake, a few days more to finish up.
