1/4 Petri dish of agar or whole dish for inoculating grain quart

So Im doing my first agar to grain transfer. I have a good amount of 3rd generation petri dishes I want to use. Some people use the whole dish, some use an 1/8 slice. Personally, I could see how using a whole dish may be unnecessary, but I havnt done it before. I was thinking about doing a 1/4 petri dish to my grain quart. Id get a lot more quarts inoculated by doing this. Thoughts?

I like to do half a dish cut into three or four pieces.
A lot of people say they don’t notice s difference in colonization time using small amounts but I definitely did. I used 1/8th of a plate and it took at least 5 days longer than normal.
One time I used two whole plates just to see and it actually wasn’t as quick as I expected. Maybe just a couple days faster than half a plate.

I. Noramlly use a single plate to inoculate 2 jars. 3-4 wedges per jar. This can vary though depending on the strain/species i grow. For example 1 plate i inoculate 4 jars with oyster mushrooms and 1 plate of panaeolus bisporus I inoculate 1 jar. Cubes 1 plate -> 2 jars
Also just a bit of advice when you cut wedges leave the outside ends as if you did have any contaminants sneak it they will most likely be on the outside wdge.

its all preference. the more inoculum you use the quicker the colonization. (most the time) I use 1 100mm dish per 4lb spawn bag. so a 1/4 or an 1/8th would be definitely suitable.

Interesting. So it sounds like Ill be doing half a dish per jar. I hear you on not using the outside “rim” or whatever. I worry that by cutting it off, ill give more chance of contaminents, no? These petri dishes are 3rd and 4th transfers, so they should be pretty decent.
How do you cut off the outside edge? I guess if you move slowly, you shouldnt have air moving anyways. I dont spray my SAB before transfers, my success rate has been great but Im thinking maby I should.

Id defenitly suggest spraying your sab and room you work in I just cut a circle then slice it like a pizza and pluck the slices out of the dish and put into jar. The outside edges you leave in the dish. Your salpel wont ever touch the edge pieces. Lucky i have a dish im about to toss.

Ok cool, you didnt let yours grow out much. Is that how you normally do it? Lastly, are your cuts sitting on your countertops?

This was a dish I was tossing so i didnt care about it didnt like the culture that was growing on it. Im isolating some strains at the moment and this one had slow growth. But i normally let it cover the whole dish. Yea its a counter but they are now in my compost pile.
