Perhaps a noobish question, but do you need to let agar come up to room temp if it's been stored in the fridge before innoc or can you just pull it out and go for it? Same question for the innoc syringes.

I wait until at LEAST 20%, you're short of 5% there... you want enough culture present to justify spreading it around... the purpose for the shake is to check the vigor /health of your culture, to give contaminants a chance to jump out ahead, and likely most importantly, colonize more evenly, quicker... shaking thus early will likely lose you a day... wait 3-4 days then mix the shit out of it

It really depends on the fruit density, but I highly suggest running heated dehydration for a full 24hrs... most dehydrators heat aro uh nd 150-165, if you have a choice,  165... the heat is believed to inhibit enzymes in the fruit which destroy the actives over time, making heated dehydration for extended period ideal for long term storage

It will need to be room temp to germinate, whether it warms up before or after inoculation

I don't see why it would be necessary. I also wouldn't bother storing plates in the fridge room temp is fine.

I don't see that fast of growth even when pouring in a shitload of LC. I think youd need to find a really fast culture for that.
I'd suggest trying out bags supplement with jars or just use jars as masters.

Might be the lazyness in me but I'd probably spawn that like it is. Sometimes I get jars that have a couple grains on top that just won't finish from being dry. I spawn as is and don't really have any problems with it.

@gizmo I don’t think it would be possible to do bag work in a Sab. Which is all I have atm.  Tho I would like to move on to bags.

Does anyone got any bag teks they recommend?

G2G is pretty quick if you're not stingy... have some patience my friend: stage projects a week apart so you always have something fruiting... that'll help

I have seen SAGARI colonize in 4 days, but it was a powerhouse culture and grain prep was on point: not typical or realistic for a goal really

You can work bags in a SAB by turning it on its side and covering the opening with saran wrap(or the lid... it's a PITA, but can be done one bag at a time

I do a qt to 10 jars and they colonize in about a week.5 given that I give a really good shaker right after inoculation. Like to the g2g and then when your sab session is finished spend a good 2 minutes on each jar, flipping it end over end and smacking the bottom corner at every 360degrees.

First, thanks for those that chipped in to let me know I can store my unused plates at room temp. You just saved me so much space in the fridge.

Two followup questions:

1) Does that mean after you PC a grain jar, you can also store it in room temp, tucked away in a sterilized tub until ready for use?

2)My PC has multiple tray layers. When I PC my agar, I found I can still fit a couple petri dishes at the top. If they are going direct to the SAB (about 3 feet away) should I still wrap them in tin foil, or can I pull them from the PC and put them directly into the SAB? Would that be too much air exposure?

Better to use grain freshly sterilized, but have seen properly sterilized bags do fine sitting at room temperature for 2wks before inoculation... this is highly dependent on moisture levels being spot on, and sterilization being proper and complete

Unless they're pp5 reusable, theyll melt... otherwise, wrapped in foil theyll not be exposed, but will need to be allowed to cool before removing to prevent spilling... making it hard to pour agar from the same vessel without exposure

So yes, wrap them lol

Once again you come to my rescue Cap!

I live at high altitude, so I already spend 4 hours per batch sterilizing my grain to be extra safe. As far as moisture, the ambient humidity up here is extremely dry. I was reading in a tek (I believe it was from someone named Bod) that dry grain works better. Not the case?

What problems can condensation cause?

As far as my dishes, they are pp5 reusable, I have a random surplus of chemistry equipment within easy access. The agar itself is actually in a capped boiling flask. I was planning on leaving them all unwrapped, let the PC finish up, and then move all of them to the SAB for pouring. I will change this up and wrap them tightly.

Man I follow eats oat prep . And every time my oats are wet and cold lol . Should I leave em till there not wet? Or fridge em till there dry?
I usually just load my jars up after 30 min dry time wet or not.. n they look wet after pc most of the time

Ideally, they should be pretty dry on the outside when you load them. At least IME. I just spread my oats out on a large screen and/or towel. Mix and toss them around every now and then to make sure all grains are getting some exposure to dry.
