So I have some 3rd and about to be 4th transfer agar. I didnt feel like growing anymore, so I just kept doing agar transfers. I couldve had like 7th generate agar transfera, but I got lazy and let them grow out the whole petri dish not realizing you could put them in the fridge.
Im not asking how to do it necessarily, but maby you could mention a tek you follow? I dont want to mess this up, as Im going to be doing a ton of quart jars. Thanks for your time and experience.
Im thinking its as simple as cutting off a big chunk in my SAB, and slowly dropping it in the quart jar but still..

Cut the plate into 8 peices like a pizza and drop a whole plate in a jar and disperse the peices around for fastest colonization time. Or you can just use one peice but itll take longer.

Good idea. I didnt think of that. I appreciate it. So after dropping it in, you cap it and shake the whole jar?

Shake it up to disperse the peices all around the jar to spread out the inoculation points. Imo it really speeds ur spawn up.

Gotta be careful how you shake at first or the wedges get stuck to the glass.
I’ve started putting both palms on the sides of the jar and moving them back and forth real quick like you’re trying to start a fire with a stick.
This kinda forces the wedges down a little and coats them in grains. Then I usually tilt the jar slightly and smack it a few times to work the wedges down.

According to my few tests, there's maybe a day difference between using one slice of that pizza and all 8 (or in my case 10). I use one slice per jar. One plate inoculates 10 jars.

You have to do it really quickly but doing 180-360 degree vertical flips of your jar is really good at getting wedges to disperse without sticking

I have a method that works for me and to my knowledge hasn't failed me in the last few years.
If I'm going to transfer ten agar plates to ten grain jars I use ten plastic tbs. spoons.
I wipe each spoon down with 70% alcohol/paper towel. I use no scalpel and flame. I transfer the whole puck in one fast motion. once the agar puck is in the grain jar, I use the edge of the plastic spoon to cut the puck into twenty to thirty pieces. I do not shake, just a slow rolling motion holding the jar horizontal with a slight tilt.. I'm sure I'll get flamed for using this unorthodox method.. Ha.
