2 Pound Bag Colonized in 6 Days After Agar!

I first wanted to say please have patients as I am new to writing on forums, I don’t know how this will look when I am done.
I just wanted to share what happened with my first agar to grain transfer.
The first thing that I did is inoculate a slant without wood with a spore syringe in a SAB and then waited for it to get huge and puffy with mycelium. (At this point I did not have a Flow Hood) (I actually did 15 tubes and only had 3 that grew mycelium and did not end up with contamination.)
I cut the fastest growing tube up into tiny pieces with sterile procedures in front of my flow hood using a scalpel and then dumped them into my bag of sterilized grain that I cut open in front of the flow hood, then sealed the bag. I then shook the bag up as evenly as I could.
* (I had used RR’s method for preparing the rye grain.)
The bag was stored at 70-82 F
I did not shake the mycelium during the colonization, I did not need to and I believe that it would have set it back if I did.
6 Days later I have a fully colonized bag of rye grain, and I am thrilled!! 😁

You basically inoculated a grain bag with a germ plate. You would have had the same success rate (maybe better) by shooting those spores directly into the grain. Its nice that you have spawn, but I think your approach was misguided.

I appreciate your observation. I may have had the same results, but the reason I did that is because I was having a lot of trouble with contamination. I think that the spore syringe had a high level of contamination in it. Instead of wasting all of the time preparing grain and waiting for it to be contaminated, I inoculated a bunch of tubes and waited to see which ones would have no contams, then I used that for assurance. I had tried many jars and a few bags before that shooting them with a spore syringe and I was tired of seeing trich. I was still confused about agar at the time too. I am near the beginning of the process of finding a good isolate now with agar plates. No contamination now that I am going agar to agar with a flow hood!

The flow hood is a game changer. Sounds like you've got the right idea. You won't know if you have a good isolate until you grow it out. Take that bag, cross your fingers for some fruits and start playing with clones.

Nah. You don't need an isolate to get growth like that. Sometimes you just get lucky with genetics.

Wow, I didn’t know that. So when I grow it out, am I just looking for the biggest one from the flush, or the fastest, or combination or what exactly should I look for to clone?
