What is growing in my agar?

10 days since MS GT inoculation, in a SAB, via flamed syringe and inoculation loop. I checked my plates today and this is the only one with any growth on it. This is my first attempt with agar, I can't conidently rule out contamination. Below are pictures lit from above and below the pastyplate.





I tried to capture the growth's dome or bubble like appearance, as if something is growing UNDER the surface of the agar. Looking back, it is possible I stuck my loop too deeply in the agar and inoculated under the surface (if that is even possible). If you have any idea what this could be I'd appreciate the knowledge. That is just some bacteria growing.

I’d shake that syringe real well and try again on a couple plates. Just a drop or so of solution and then spread a Z with your loop or just tilt the plate. You will often have bacteria or other stuff in a syringe so that’s ok but you want myc to be able to grow out past the edge of it and not be enveloped. The swipe pattern helps with that.

My fears confirmed. Thank you! If I haven't seen any myc growth at all in ten days should I chuck the plates and start everything fresh?

I’d still keep them for a bit just because I’m a hoarder of those things and you never know what might pop up. But I’d also start some new ones. Only recently have I had a print not germinate on agar, I swiped another spot on the plate though and got growth that time. I’ll usually see something on day 2-3 if you hold it up to the light...but be careful if using syringe water that might still move around.

Nothing else has germinating, tried inoculating more plates today with the same syringe. Here's a pic of the growth for curious

Is that fuzzy good myc I see on top of that bacteria spot? If so you can scrape up some hyphae and swipe them onto a new plate. Using an inoculation loop or a scraping motion with a scalpel.

Agar is great man. There’s no such thing as contaminated agar (unless it wasn’t sterilized and the whole thing is full of growing junk) just cultures that need cleaning. So you scrape some of that good stuff off and move it to a new plate or even just to the other side of that plate. Probably you’ll still bring a little bacteria but this time you should have more myc growing outside the bacterial zone. Grab some of that to a new plate and quite possibly have clean myc and if not you try another transfer.

Because you have no other growth on other plates And this one is only that spot, at least you know your sterile tek has been ok. Otherwise you’d have little satellite colonies of mold and stuff popping up in places where you didn’t inoculate.

Thanks! I'm gonna try this today. I have a loop and a scalpel so I'll try both, at least for the sake of practice. I can't really tell what's growing there it's like nothing I've seen in any pictures, but I'm gonna try to transfer some today. Inoculated a few more plates yesterday with a loop and syringe, let's hope for some growth!  :alien:

Sweet. It definitely looks like cube myc from what I can see. It’s growing on top of bacteria so that will affect how it looks and grows but if it is good stuff you will know in just a couple days. Sending good vibes :mushroom2:

Okay, small update/round 2 of What is Growing in my Agar?!

I inoculated 4 plates from the obviously contaminated donor plate. And the donor plate is clearly some kind of mold. Here is a picture of it today:


And a picture of the only plate that is showing growth:


A little hard to tell just how green the growth is in the first pic but it's green. And the little dark spot in the foreground of the second plate is also green.

So... did I just streak a bunch of trich thinking it was cube mycelium or is there anything worth saving? My MS to agar experiments have not been successful so far, hoping that when I can actually get a print and can stop using syringes that I'll have a little more success.

It looks like maybe that wasn’t cube myc sitting on top of that lump after all. It’s still really tough to tell from the pics exactly what is going on in there. Petri dishes are the bees knees.

I can pretty much guarantee you’ll have better results working from a print. I’ve only had one print swipe not germinate and when they do germinate I almost never see any obvious contaminant spores germinating. Whereas with syringes I’ve had something funky show up with each of the 6-7 I’ve used on agar.
