Germination still possible?

I had a sterile medical cutip swab and tried using it to streak my agar plate. It was easy to get the spores on the cutip from the print, but when I went to streak my plate it appeared nothing came off. Everything was still on the cutip and not one black speck came off. Then I grabbed another plate and funneled all the broken pieces and loose bits of spores from the print and dumped them all on another plate. Will these spores still germinate if just dumped onto agar? Or it needs to be streaked and spread? The only reason I did it a second time this way is because nothing came off from my swab the first time, it looks like I didn’t even streak the plate, so now I have a second plate with a bunch of loose bits of spores. Will germination still happen like this?

Chances are the swab worked. Dumping spores onto plates will work....but such a waste :sad:

Dumping the spores from the print or anywhere else will for sure work. I've used Q tips before, but I find that just scraping a tiny bit with the scalpel and dumping them on the plate works way better. Im new too and I always make sure I see black chunks in the agar. It might be wasteful, but I like the peace of mind knowing I for sure put enough spores down.

Spores are microscopic alot of times you won't really see anything off the swab until germination.

Yeah I know, but I guess it's just a peace of mind thing actually seeing visible confirmation that it is on the agar. Thanks! Last time the AA+ took exactly one week to germinate. I'm trying out Burma now, so hopefully germination occurs.

I've been working under the impression that more Cube spores on a print or more liquid from a MS syringe also means more contaminant spores intermixed.

It can mean that yes. We use agar so thats not really a problem. Sometimes less is more. I guess no harm no foul though right? Lmao.

It always seemed like I had a much better success percentage without contams with MS inoculation of PF jars when running 25-30 jars per 10CC syringe. Maybe it was purely luck, or perhaps my sterile technique improved from when I was making 10 jars to 10CC.

With PF Tek you are definitely better going very light on spores but when using agar it's not as much of a problem since the good will be isolated from the bad.

Very true, it's just become habitual now to try and use as little as possible when making petris.
