First transfer wedge selection help

I have finally succeeded transferring a tissue culture to agar!  It is now time to make some transfers in an effort to isolate.  I would love to hear your opinions on the best positions to target for taking wedges.

at this point, just pick a spot that you like and go for it. make the transfer as small as possible. I also only recently learned, that further isolation from a clone tissue might not desirable in most cases. You really just want to get a clean, nice looking plate, which you seem to have.

What i mean by "learned" is, that some TC wrote it in some thread. I haven't really looked into it further yet.

The reason is that as you isolate more, you may be isolating traits you dont want.

If you have alot of different genetics then its really just a 100% clean ms inoculation.

That almost looks like an isolate already. If you're set on getting a monoculture, you should probably wait a few more days to get some sectoring and then transfer whatever you feel is looking like a desirable growth.

I'd first make a transfer to another plate and a slant to keep your master that you can return to at any time.

Isolating is hard work with possibly a lot of undesirable results.

Yea I say next plates are good for inoculating some grains as long as they look good still.

Thanks for the feedback. 

So further isolation is not really needed but I should still do one transfer just to ensure I have clean mycelium before going to grain?

Also, I don't have any slants, how long can I save a colonized agar dish in the refrigerator?
