I see bacterial colonies on almost all dishes and it's no surprise due to the amount of water there is inside the 'petri plates' what kind of containers are those anyway? What are the dimensions looks more like a 1/2 pint jar to me. Also did you take pics of the cultures with the container lid off? Only time you should open the lid is when you need to open them.


Also no one knows how a culture will perform by I cpecting it visually I side a petri dish, you'll know that only after you fruit it secessully over and over using different methods, only then will you start to form a picture of how that culture performs, so instead just focus on making agar dishes with no condensation and take a small transfer from pretty much anywhere you want to begin with because you can't count on the culture being clean anytime soon, you'll definitely need a few more transfers and then again the transfers need to me made into receiving agsri dishes that don't have water inside them.

Those are glass petris that you first poured and then sterilized? Yeah condensation will not be avoidable that way, but if you start pouring agar you'll avoid the condensation. Also pressure cooker is not really needed to sterilize liquid media's (like agar) tho I recommend using it if you have it available but simply boiling liquids will sterilize it :thumbup:

That doesn't sound very methodical. Your pouring temperature should be between 45-47C not warmer not colder than that.

You mean from what ketchup cup? :lol: it doesn't matter, they're all contaminated just focus on staying away from the outer edges cause that's where the bacterial colonies are.

Bacterial colonies are those vaguely and differently discolored parts throughout every 'ketchup cup' and basically everywhere spread across the entire surface of the plates. I means cups. Some cups have bacterial colonies mostly around the edges but it's very obvious and can be seen clearly. Plates should look something like this:

You are using ketchup cups instead of plates?
Have these cups been sterilized prior to pouring?


Also, don't worry so much about strains right now. I'd focus on getting a clean culture to inoculate your spawn with, and cloning fruits.
Strain isolation is a very long and involved process.

As mentioned, dont worry about strain isolation. All your 3rd transfer plates look clean to me. Id use any of those third transfers :thumbup:

I only see bacteria on pics 1-3. I think it's some weird glare on some others.

Look up 2 oz slime cups on amazon (pics for reference). They are like 15 bucks for 40. You can poke small hole in the side with a hot screwdriver and put mp tape over it. The condensation will work it's way out slowely.  They are a bit larger than the catsup cups but you can pc them and reuse them. I have had good luck with no poor. While having real dishes seems cool and super useful to grow larger wedges, the poor is another time you can contaminate if you are still perfecting your steral technique. I let them sit for about a week so I know they are clean before inoculating them. It's also helpful to rotate colors as it helps keep track which dishes are older. 

While your A plates dont look so great, you could probably get some good transfers from the B T1 plates. All three have some decent looking edges between 3 and 6 o'clock. At least from what I can see.

Roughly how deep are those? And across the top?
Those look pretty great for no pour plates  ! I prefer pouring plates but I hate the waste.
I used Pasty Plates for a while but I dislike not being able to view thru the lid.

Also do you not have to wrap them in foil to PC them like Pasty plates?

I put a small hole with mp tape on the side near the mouth of the plate instead of on top and then dont need to use foil.  I cant remember where I got the idea, either in pasty's thread or in the holy grail Mlno pure tek. Each plate gets about 2-3 tablespoons and ends up being an eighth to a quarter of an inch. The plates are 1.18 inches deep according to the vendor.

I dont care for the condensation either but I usually look through the sides. Also using lme agar makes the plates clear enough that I can look through the agar with a light on the lid.

I like A1 and B3, Im new too, but this looks pretty good. when You transfer just keep away from the grayish shit on the edges.

This is probably the reason why there is bacteria in all the 'ketchup cups' :lol:
Just take a tiny transfer from inbetween the center and the edges of the cup (right in the middle of the culture as far away from bacterial edges and bacterial center as possible, at least for this transfer) and of course your receiving plates or cups should be properly sterilized :super:

You may be able to pc the cups. My understanding is it's the lids that dont always make it
