Agar to rye ==> are these plates ready?

Hey guys,

I have 6 plates looking clean on a first transfer from this B+ clone:

I'm planning on doing a second transfer tonight from 2 of the plates, but I'm getting pretty antsy to start my next grow. Are these 4 plates far enough along to transfer to grain?



I also have a bunch of B+ plates on their second transfer from a MSS. I've read that its a good idea to wait for MS agar plates to start pinning before transferring to grain to make sure they will actually fruit. But again...getting kinda antsy. I'll do a 3rd transfer on a few of these plates tonight, but if I get a possible green light to transfer the above clone plates to grain, I might transfer some of these MS plates as well. Or am I jumping the gun on that?

Those look good. I think it would be worth transfering one of those petris to grain and see what happens.

Don't know where you read this.

You can inoculate grains with agar as soon as you are sure you have a clean culture. I grow my plates out about 3/4 of the way and use 1/4 plate per jar. But you can inoculate with more or less, it's preference.

Another quick question...

I'm trying to segregate plates that I know are clean from those that are slightly contaminated. Can anyone confirm that mycelium will change the color of agar during colonization?

I have several clean looking plates that the agar has shifted from food coloring blue to more clear, and in some cases reddish from the bottom view. Second pic is bottom


I plan to transfer contaminated plates to new agar, and transfer fully clean plates to grain

That culture looks clean. Zoom in at 3'oclock though, you see that tiny yellowish blob? That's bacteria.

Sweet. Double checked that plate, and the blob was stuck to the outside surface of the lid.

Thanks for confirmation. On to transferring dirty plates
