Hey guys. I have a some problematic agars going on here. 1) Why is the mycelium growing up like that in all of those agars? They are the fifth or sixth transfer by now. 2) I had some previous agars couple weeks ago that had the same orange discoloration underneath the transfer sample that can be seen at picture 2. What does it mean? Is it bacteria? 3) What could be the possible contaminant at picture 3?
Thank you a lot

Looks like 3 is toast regardless by the looks of those satellites. Whats the agar recipe? nute levels being way off can cause some weird growth.

I forgot to tell the recipe. It's the common malt extract recipe (20g agar, 20g LME, food colorant, 1l water). I have some other agars with the same recipe growing up fine.

I don't think you have any mushroom mycellium there at all, but I could be wrong, could be some wonk agar causing some uber weak growth. The discolouration occurs as organisms digest into the agar, they digest the food colouring.
I liked LME a lot when I started but it started giving me weak growth so I switched to PDA. Maybe there is some issue with LME over time as it absorbs moisture? Certainly if you use a constant weight of LME and it is absorbing water over time, it will have less and less nutrition per weight. I wouldn't think it would confer a result this substantive though. I think your plates are prolly just garbage.
I've been quite liking 750ml broth from one small red potato boiled for 30 min + 15g dextrose lately.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister. I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave. I think it will be good cause B+ is so potent. Triggered yet? Only a square would say "a cube is a cube."
No, this does not look right...

LME needs to kept in a mason jar or it definitely absorbs moisture. My first batch got as hard as a rock and eventually I couldn't even take shavings off it. In a mason jar it stays just like new.

Maybe I'll give that a shot, I had it double backed with desiccant, but LME is better desiccant than silicon. I never did it cause I knew if it did turn into a rock in that mason I'd have to throw it away cause I'd never get it out.

All right. Already threw those plates anyway. So the discoloration is a normal process then? I was concerned about it because some plates had it and some others didn't. And now that I recall .. I had some plates that the discoloration area way overreached the mycelium growth, I mean it went further than the fungal growth (although the one shown is a good example too). That now leaves me in doubt now if it really is because of the myc

Usually the digestion of the food colouring will be about ~1/4"??? trailing the leading edge of mycellial growth. I believe this is because it grows over the surface of the agar and then into it over time. If there is evidence of digestion of the food colouring ahead of the mycellium, that is a bad sign. Possibly indicates insufficient sterilization or bad bad filters if you see no surface contams but that kind of discolouration.
