Help me understand what is happening on my agar
    #26652512 - 05/06/20 08:49 AM (3 years, 9 months ago)

So I’ve just made my second agar to agar transfer. I’m still not sure about what kind of growth I’m looking at. Where the best places to cut a wedge from is? I always thought contams would be easy to spot, probably some nasty green looking stuff but now I wonder about some of these translucent spots like in these pictures below. Most of my plates from transfer 1 look really clean, no growth outside of the main myc growth. These two I decided to scrap due to the weird blotchy stuff away from the main growth. So being as I was able to pull the tops and get some decent pics I wanted to post them and see if someone can just walk me through the types of myc growth and whether the questionable spots are actually contams. Where would you take a wedge from? Still just the leading edge? Thanks for any help or critiques

I would want to hear others opinions on this as well.

If these were my plates, I really like the 12 o clock area on the 2nd pic

The 5 o clock area on the first pic looks decent too.

These areas look healthiest to me, with my limited experience

Depending on how clumsy you've been when placing your transfer to a new plate, it could be myc. That thing at 3 o'clock on the 2nd picture is definitely a contam.

I'd take transfers from 5 o'clock on 1st picture and 1 o'clock on the second asap.

It was clumsy. I’m having a hard time getting the wedge off the scalpel. When I do I usually have to move the wedge to the center of the new plate.

I've had those problems as well, but turning the scalpel upside down and kind of pressing it into the agar has helped a lot, now I get it off in one smooth move first try.

Nice, I’ll have to give that a try.

in general just pressing into the agar can help, either side up. no need to be afriad og cutting the agar. i think i do it upside down most of the time tho.
