The 12 shoeboxes experiment 2

Since I am stuck home and have cravings for mycology, I have decided to start an experiment...
The experiment is about growing several varieties of Psilocybe Cubensis, within the same conditions and see if their fruits end up with specific characteristics or if they all just look alike and "a cube is a cube". I might also select a few fruit for cloning if some of them look interesting !
For each of the varieties, I will follow this exact procedure:
Quote:
Spore print > Germinate on agar plate > Transfer on new plate > Second transfer if needed > Inoculate grain spawn > Spawn to bulk in the shoe boxes > Grow fruits
Here is a list of my equipment:
- 12 Jars
- 12 Shoe boxes
- Blue agar plates (MEA)
- Spores Prints
- Popcorn grain (can't manage to get anything else right now)
- Coir or CVG, not sure yet
- SAB
- Label printer
Since I have 12 shoe boxes, this is the limit of varieties I will grow side by side.
So far I started some agar work: Already transferred a wedge to a new plate
- Golden Teacher
- B+
- Brazilian
- Lizard King
- Thai Elephant Dung
- Mazatapec (second transfer)
Germinating:
In the upcoming weeks I am expecting those varieties and may receive some extras even
- Creeper
- PESA
- Malabar
- McKennaii
If I don't receive them on time, I will just put the current plates in the fridge to slow down growth.
Look at these boxes waiting to fulfill their destiny !!!

In my experiences with different varieties, they do have different appearances, while not overly significant Unless we’re talking about albino varieties or PE. Those are, obviously, way different looking.
Typically when we use the “cube is a cube,” we’re referring to growth conditions and potency. Except PE, again the anomaly.

Good news my friends!
I woke up this morning and found that my mail box was full of spores ! Not the ones announced in my first post, these seems to have been lost or delayed...
The KSSS spores were bunk, so this strains is out of the grow.
Here is the final list:
- Golden Teacher
- B+
- Brazilian
- Lizard King
- Thai Elephant Dung
- Mazatapec
- Z strain
Plus the ones I will germinated right after posting this :
- Matias Romero
- Avery's Albino (supposed to be true albinos !)
- Tidal Wave (F3, so unstable they said !)
- PE6
- Leucistic Cambodian

My shroomerites, here are the 12 jars, I just inoculated 2 minutes ago

You may notice on the pic that the KSSS is back in ride ! The spores germinated later so I decided to take it back
From front to back: - Brazilian, Golden Teacher, KSSS, Matias Romero - Mazatapec, Thai Elephant dunk, Tidal Wave, Z Strain - B+, Lizard King, PE6, Leucistic Cambodian.
This time, I did use a mix of popcorn, gypsum and some coffee too.

If you are going from spores, I bet you could do 12 boxes of the same variety and most of them would have AT LEAST slight differences
There is a huge amount of genetic variation in a spore print

Well... Things don't look good so far. It's almost two weeks after inoculation and I have only 6 jars that seems to colonize very slowly, they did barely colonize 15% of the grains. The other one have literally colonized 2 or 3 grains and then they just stopped... Should I throw these ?
The jars are sitting on the table and room temp is 21°C/70°F. I think the temps shouldn't be the problem here.
Maybe a gas exchange issue? I am using micro-pore tape, one layer in the inside of the jar lid and two layers on the top.
Also, I have a cheap PC, it's only reaching 11psi but I always cook my jars for 1h30.

I'm cooking my jars for 1.5-2 hours at 15psi usually. Your time was way too short imo for 11psi.
It's not a GE issue if you only have 15% colonized. I'm betting your grain went sour.

I tossed the 6 jars that did not colonize more than two grains. They did not smell at all, it just smells like corn.
BUT the kernels are very hard, I am not even able to break them with my fingers.
I never soak my corn, I just pressure cook it for 30mins to hydrate it and then load it in my jars.
I read in some other popcorn tek to soak it for 24h and then just simmer instead of PCing. I might want to try that and also maybe PC my jars for a longer time.

Yeah the added coffee upped the nutrient load.
Really agar is the best way to get clean cultures and healthy spawn.
Start over and try again!

Are you using popcorn or he
Dehydrated deer corn?
Popcorn shouldn't need a 24 he soak because it still has some moisture in the kernel when dry.

I am using normal corn, the one you can cook on a pan to make popcorn.
I will try to make things simpler next time: No coffee with my corn and work with only one variety until I manage to get a successful grow with grain spawn.
