Snake Venom *vs* Chitinase
    #26648853 - 05/04/20 06:26 PM (3 years, 8 months ago)

Reading Alan’s thread today about this holotype collection of Cubensis and it’s lineage (which was most enjoyable, check it out here in Hunting: https://www.shroomery.org/forums/showflat.php/Number/26647546/page/1), I heard mention of Snake Venom for hybridizing between closely related species and was reminded of an idea I had long ago, when I first read about Snake Venom being used for this purpose: Chitinase in e.g. species like Coprinus Comatus which deliquesce at maturity. Of course, it would be amusing if we could use mushrooms to hybridize mushrooms.

What is special about snake venom that allows it to work for this purpose - from what I’ve read, to weaken the mycelial cell walls. Is it that it will survive heat treatment by agar?

Theoretically, could one simply collect the ink resulting from the caps deliquescing and add it to an agar mixture similar to snake venom?

Also, I found this nice short article on the subject: https://blog.mycology.cornell.edu/2008/07/01/the-dish-on-deliquescence-in-coprinus-species/, if you’re interested.

Someone correct me I'm wrong, but I thought snake venom had to be added after autoclaving when the agar has cooled down a bit.

Interesting link about the comatus mushrooms. Interesting though it says the chitinase it makes doesn't affect the stipes or the mycelium, only the caps. So I don't know if that specific chitinase would work on mycelium on agar.

I believe the snake venom method is bunk. Check this thread for more information.

Thanks Pinback, that is an interesting thread! It certainly appears you are correct, but I should read the paper myself of course.

As for Chitinase, quoth drake89 in your thread:

All the papers i have read use chitinase to form protoplasts .

So - looks like I have more research to do. I’ll try to gather some papers here in the thread as I go.
