    #12039606 - 02/16/10 12:12 PM (14 years, 10 months ago)

Just wondering how many wedges of agar you put into a grain jar.
How many wedges do you use
You may choose only one

Bumping this up after 10 years, because I want to know what you guys think in 2020. Is one wedge really enough? Is the benefit significant enough to transfer more than one piece of agar to grain?

Lol 2009

It's all preference. I like to make my inoculations quick, so I use about a 1/4 plate per qt, drop it and close the lid real smoothly.

Thanks, I'll go ahead and use one wedge to only have to expose the sterile grain once in my SAB.

I use a single quarter-plate wedge per jar. If I inoculate with multiple small wedges they usually stick together and end up having less surface area.

More agar means more myc. I rarely use less than a quarter plate, usually half a plate cut into a few pieces.

I’ve used two whole plates before when I had way too many of a clone culture sitting around, definitely went fast.
