Question about grain to grain transfer

Just before I start, I did search my question first and I didn’t come up with anything, so here it goes. Around end December, or first of January I cloned a really nice PE specimen, I did a agar to grain transfer and from there I have been doing grain to grain transfer from it since. My question is how long can I do g2g transfers on this batch before there are any issues. Any insight would be greatly appreciated.

Ive done it like 4 times i think. Maybe more
Make more agar and use agar

Most dunt go further than three or four. There isn't really a scientifically verified answer to you question because it entirely depends on how much grain/ myc is being transferred to each bag/ jar. If you stretched 1 jar to two new jars the cell division would be way less than stretching one jar to 18 4 quart bags (yes, you can do that).
Another reason is that most don't push the limits because every generation is further and further from your verifiably clean culture. Meaning, the more generations deep you get the more vectors you've had.
Also, if you follow the good old advice, 1 becomes 10... These 10 become 100... Those 100 become 1000. So, your three generations in and have a thousand jars. Obviously you don't need more than that.

I don’t have the original plates, I’m running low on alcohol so I figure g2g transfer would save on supplies.

Gotcha. Im probably at 5 with this batch. It may be my imagination but I have noticed that this recent patch doesn’t have the vigor the last generation did. As some as I can get my hands on some alcohol I’m going to start some new plates.

How do plates require less ISO than G2G? The same exact principles are in play.

G2G requires less ISO/amount of inoculate I guess.
