How to get spores into agar
    #26635862 - 04/29/20 02:31 AM (4 years, 8 months ago)

Very new, planning my first grow.
This seems like a simple question, but I've looked around and I've not found something that really answers my question.

What are the ways to go from a spore print, to inoculated agar? Obviously there's the spore syringe, however I'm wondering whether there are any other notable methods that are practical, better for certain things, or easier. I'm just trying to get a lay of the land before I commit.

An inoculation loop (can be rigged up with some wire and an exacto blade's handle) or just the blade can work. You will want to flame sterilize your tool and cool it in your workspace before carefully scraping up a bit of spores from a print.
There's a handful of teks from trusted cultivators that talk about this, so make sure you use that search tool to narrow things down to the recent and relevant. (Tip: Check the "Trusted Cultivators" box and stick to posts newer than 5 years)

Scrapping spores off a spore print using a small metal tool while in a SAB is probably the easiest way

Spore syringes by their nature are inherently dirty, because even if you use an indoor fruit to spore print it's still been exposed.

Like these guys said, use an inoculation loop. Flame sterilize it, then cool it in agar in your recieving dish. Rub it in tiny circles to pick spores up off the print.

Swipe them across the plate in a zig zag pattern. This makes it easier to identify clean germination.
After germination transfer clean myc to a new plate

My favorite way is the gif in my sig, works great even for dirty prints and makes it way faster to get clean , well organized cultures vs a standard swipe

Detailed in the agar guide in my sig as well

Use a flame sterilized knife tip, metal inoculation loop (diy one from a stripped bread tie twisted around a toothpick) , a sterile cotton swab(6 inch medical grade ones work great. There is also "spore dilution " and "serial dilution" which involves scraping spores into sterile water and then using ,pipette, syringe, inoculation loop , to place a drop of the diluted spore solution onto germination media.

Here is another good one, cardboard germination.  This would be done with hydrated sterile corragated cardboard, either cut into rounds and placed in jars or petri dishes and swabbed with spores.  Or a cool one, using a small tube or jar and a paper punch , knocking a dozen confetti peices out of cardboard and hydrate them in a tiny bit of malt extract.  Sterilize them in the jar or tube, using a hemostat sterile tool to grab one and swab the print and place a few of those into the awaiting sterile jar or tube filled with a bunch of other confetti cardboard peices. It will germinate and you can simply grab one and place onto agar.
This method was taken from a book that I have linked below.

Although some methods are preferable over others in w felt the need to put all the ones that immediately come to mind.

Easy peezy bro. Just lick your pinky finger and lightly dab it on the edge of the spore print. Then just streak your pinky finger tip across your glass peetree dishes. Welcome friend.
