Scratch and Drip Method for Spore Prints


This Tek was inspired by Josex's write up on the biopsy method. The idea is to use less spores to reduce the chances of contamination. The germination step is critical and it is important to be sure you are off to a good start. If you are starting with a print, I recommend this Tek.

What you need
1 clean 20ga luer lock syringe filled
Flame for needle
Spore print
SAB
Agar

1.) Organize your SAB or flowhood area and make sure you are ready for action

2.) Get your clean agar plate ready for the inoculation by cracking the lid and resting it so that you can make a quick slip and drip

3.) Open your print and have this ready as well before you begin to flame your needle

4.) Flame needle outside of your box and move it inside

5.) Now that your hot needle is in the box and ready for the scratch, release a small drip to cool the tip (remember to do this inside your box near your print)and flick the remaining drops off to make sure your tip is not soaked

6.) Now you are ready for the scratch. Simply scratch the edge of your print with a couple of slight strokes to pick up a small amount of spores

7.) Crack the lid and release one drop into the plate and quickly close

8.) Secure lid and distribute liquid by moving the plate around or just leave it be.

Germination will take the average time of 7-10 days or so. Here's a look at my pe plates inoculated with the scratch and drip. Made 12 plates, all germinated and one contaminated. This is from a print I recently had trouble with swab inoculating.

why not just use the loop? you also work with a very small amount of spores that way

Loops are cool but I'm using a needle tip instead. I'm picking up less. Plus I like that I can sneak it in through the crack of my no pour lid.

I think I've tried something like this on a foil spore print and tore it which is why I started using the loop.

I don't mean to knock your Tek but the downside I see to not using a loop is not being able to swipe the plate.
Swiping spores in a zig zag pattern across the plate really makes a big difference when it comes to transfering clean myc after germination.

Your good bro. I agree with you but I'm getting some beautiful germ plates, especially with the pan species. I usually do not see growth like that until I've transferred several times.

As for the scratching, I've done it on a few prints now without any damage. I'm just barely touching it.
