2nd or 3rd generation of isolates (though honestly I probably wasn't isolating correctly, as the growth still looks very uneven). I'd imagine the two on the left are the best looking, but I'm not sure if I should nocc with them yet. Thoughts?

You could but I would take a transfer from any of the rhizomorphic growth to try and further isolate.

You could do both if you're really hankering to get this variety on grain!

i.e. take a couple slices from a sector to make a shoebox with, then utilize other sectors for further isolation :cool:

And then if you end up with something awesome in your shoebox, you could clone from there :smile:

How constrained should an isolate be? If you look closely in the picture you can see roughly how big the sections I took from the previous plates are. Is it normal for 3rd generation to be this varied in terms of growth? Or was the isolate I took too big?

Those are rather big transfers for strain isolation you only need a very small fragment. It can take a lot of transfers to get an isolate.

check out this video


I agree with fungusam. I find it best to go both routes since getting an isolate can take so long and there is no guarantee it will be a strong fruiting strain. You will have to test each strain separately. The nice thing about a ms/multistrain grow is you can test a ton at once clone and then isolate if necessary farther.

best to not put all your mushrooms in one basket
