I recently ordered this cordless Kitchen Aid blender. I look forward to taking it for a spin, so to speak.

That’s a nice stick blender, I use the shit out of mine. I also make hot sauce though.

D3monic,
You might have mentioned it before, but can you please remind us how much activated charcoal you use to create that sexy black agar?

I don’t use activated. This is the brand I use Organic Charcoal Powder, 1 lb -... https://www.amazon.com/dp/B07DWCMRQ6?ref=ppx_pop_mob_ap_share
Good heaping spoonful per media bottle

What size media bottle? Or rather, how much charcoal per how much agar/MEA solution?
Do you just dump the charcoal into the media jar and slosh it around with the agar/MEA solution to mix it? Could the charcoal be added to the agar/MEA solution while it is still heating in the pan and being mixed?
Thanks.

I don’t see why not but that would be rough on the blades if using a stick blender. I just add my food coloring or charcoal to the media bottle then funnel hot agar into bottle that’s enough to throughly mix it. Def don’t shake or anything more than a gentle swirl as this will leave persistent air bubbles.
I split 1000ml between 3 media bottles

I weighed it out, basically a gram per 100ml of charcoal. Was 3.5g scoop for ~350ml

Food coloring has alcohol in it? Or which one do you use?

I presume you mean one gram of charcoal per 100ml of agar.
Thanks for the numbers.

Thanks for that info, D3.
Would you also share with us any dope color mixing combos you've found w/ the food coloring you linked? I have both sets that are available on amazon from Wilton.
Also, have you tried any other Wilton products? Like, does the Icing Color work the same as the Food Color? Here's their icing color set: https://www.amazon.com/Wilton-Master-Icing-Color-1-Ounce/dp/B004EBVLBS/ref=sr_1_5?dchild=1&keywords=wilton+color&qid=1595628413&s=home-garden&sr=1-5

Haven’t tried any of their others and after some time I’m starting to lean away from their magenta. I get better results on the blue and orange/amber colors.

Thank you D3 for such a beautiful tek.
If I may ask, when doing transfers, do you punch right through the petri caps (meaning do you open the petris and only cut the myc on agar, or do you keep them closed and kind of melt through the cap of the petris)? Asking because can't see clearly on you photos, am blind sorry
Also, using food coloring when working with agar is purely cosmetic improvement, or are there any other reasons behind it?
Thanks!

Purely cosmetic, though there’s some contrast benefits to aid in identifying contamination. Def remove the lid and punch through the agar. Tek works best in front of a flowhood where you have freedom of movement but is totally workable in a SAB as well.

I had a whopping 3 transfers under my belt before I found this tek and was immediately hooked.
That first picture absolutely captured me.
A week ago I made 9 swift transfers and tonight they’re all beautiful. With that circle array it’s so easy to spot the healthier mycelium.
I poured 30 plates last night using your recipe. I swapped grain water for your LME, I used distilled, am lucky enough to already have that amazing immersion blender, Amazon’d the food coloring, and tonight I’ll take those 9 transfers to T3.
I have 15 of the light blue, they turned out sexy. Definitely key is taking extra care to get all the chunks with the blender. I made sure it was as homogenous as possible.
I used your water bath method and though I got a small bit of condensation, only a few of them still have any at all, and most of them are crystal clear.
Those blue plates are so sexy I’ll definitely post pics of them post transfer 👍
I think back to the hack job of a transfer those first 3 were 🤣 I can’t imagine doing it any other way.
Although a tool like you mentioned that is a touch easier to wield could be useful.
Thanks for a great one D3 👍👍

Pics of tonight’s work with using D3’s Tek:
Also made a lemon lime color.
This first pic was a transfer using the D3 punch method, but on my grainwater agar. The large cultures with multiple transfers taken were 2 of my original 3 first ever transfers, done with a scalpel.

Is distilled water much better than regular boiled tap water? Is it worth the investment?

Yes in my opinion it’s worth the $1 investment. Purely anecdotal but I’ve noticed a pretty good change in growth between tap and distilled. Of course everyone’s tap water will be different.

Yo D3monic,
Have you noticed any differences in growth (Magenta aside) between non-colored plates and colored? I checked the Wilton box of neon color dyes and they all contain corn syrup & glycerin. I'm wondering if this alters the nutrient profile of our plates at all in any meaningful way?
The primary color set by Wilton, on the other hand, contains sorbitol & glycerin instead of corn syrup as a sweetener. Curious about that as well.
