I tried the emulsifier blender with this pasty recipe. It’s a little tough but I think it made the plate mix smoother. I put dark purple food coloring so it’s tough to see. I didn’t like the all metal tool. I need some kind of holder so it doesn’t burn my hand. And I almost set my table on fire tonight using a paper towel to hold it while flame sterilizing the utensil. Yep noob here. Anyway, I used the smaller tool this time and I like the size. I used a bigger tool last time and had some difficulty.

Awesome eats, yea.. def can’t heat till red hot. Will be hot as ish and take forever to cool off. I flamed in the side hole out the tip for 3-4 sec or so then roll the tip in it a few seconds. The new tool I have is brass with acrylic handle. The brass gets red hot almost instantly.

When I’ve been looking at the leather punches I’ve been thinking of wrapping the handle end with some silicone fusion tape. It’s not going to be as aesthetically appealing or lasting as a proper custom handle but it should insulate from heat...maybe

Plumbing or electricians sections of the hardware store I think. It stretches and sticks to itself without adhesive. I use a strip to secure tyvek over the neck of my media flask for the PC cycle. Just looked it up on the zon. I forgot how expensive it is. Averages $10 for 10’.

The punches arrived. I used the 5/16" punch because I wanted a slightly larger puck to manipulate on my first attempt.
After sterilizing and cooling, I punched the plate and it cut a perfect puck. On the second punch, the puck stuck in the punch. This was my first agar pour and my agar is deep. I poked out the puck with the scalpel and left it on the plate. I punched another puck and it stayed on the plate.
I speared the edge of one puck with the scalpel and transferred it to the plate.
I somehow managed to slide the scalpel under the second puck and lifted it off the tray. It was time to load the slant.
Instead of turning the scalpel upside down, hoping the puck would stick to it while I inserted it into the slant, I inserted the scalpel, with the puck balanced on top, into the horizontal slant. I rotated the slant on its horizontal axis so that the agar was above the scalpel and puck. I raised the scalpel, sticking the puck to the agar. Then I rotated the slant and scalpel on its horizontal axis again so that the agar was back on the bottom. I slid the scalpel off the puck without a problem.
I'll let them grow a bit and then into the fridge they go for long-term storage.

I also bought your recommended agar, malt extract and food coloring. I'll use them for my next pour.
Thanks again.

The punch is great! You'll quickly learn how to effortlessly slide your scalpel under the plug, turn it face-down and transfer it to a new plate.
Thanks again D3monic!

Yeah i love it. Now, i think i prefer the smaller size hole punch better as i can take a smaller sample of tissue.

1/4” works good. What size you using?

The 1/4" punch is the way to go. It's smaller diameter minimizes the likelihood of transferring potential contaminants from the surrounding agar.
Try it. You'll find that it's easy to slide your scalpel beneath the plug and transfer it.
Good luck!

D3monic,
After growing them and doing my best to prevent contamination, I want to transfer some plugs to 125mm master slant test tubes with 7.5ml of agar and wood.
I've purchased and plan to use your recommended agar and LME for my next pour.
Is the 20 minute PC time still appropriate, or do slants require less time? Ideally, I'd like to PC both the media bottles and the slants in one run.
Please advise.
Thanks again,
Tom

D3monic,
After dunking the punch in iso and flame-sterilizing it, sometimes what appears to be residual, burnt carbon is present in the punch and gets transferred to the agar plug. It can leave black discoloration on the plug.
I don't think a bit of carbon on the agar will hurt growth, but it causes the transfers to look "dirty" and gives me something else to worry about.
I'm striving to create plates as beautiful as yours.
Have you encountered this phenomenon? Aesthetic considerations aside, should we worry about the "carbon issue"?
Please advise.
Thanks,
Tom

Once in a while I use a iso soaked paper towel and ream out the inside to get carbon out. The build up depends on your flame source. I get very little to none

I'm using a butane torch.
I'll ream the punch with an iso-soaked cloth before sterilizing it. A paper towel might leave bits of paper inside the punch, leading to more burnt carbon after the next flame-sterilization.
Back to my original question. Have these scant carbon deposits negatively affected the growth or health of your mycelium?
Thanks.

Not that I’ve noticed in any way.. I mean I even use carbon powder in my agar to make it black
