Tom what brand / color you using? I typically use at least 2 drops some times 3. I wonder if some the brands have a shelf life and start to break down or if ingredients separate in the bottle. Maybe a good shake before each use.

D3,
I used your recommended brand of dye, in this case the blue. I shook it before using it. In your original post, you said that you used one drop of dye per media bottle. Thus far, that recipe is a proven winner.
Have you changed your recipe? Please advise us.
Thanks,
Tom

I use 1 drop per 100ml in the bottle. Though 2 drops still gives a vibrant color up to 300-400ml not an exact science and havenโt noticed any issues with more or less.

I used three drops in 300ml. That was clearly too much.
I'm not a chemist, but there must be sugars or other compounds in the dye that can potentially over-drive the nutrients and produce these results. I'm sticking to one drop.
Thanks.

Possibly, I wonder if perhaps the corn sugars are fermenting into alcohols maybe?

Could be. But I have no interest in testing the hypothesis. I'm sticking to your original tek.

A word of warning. Do not use more than one drop of dye per media bottle. Much like tripping, I fell in love with the colors. I put 3 drops of blue dye into a bottle and poured plates with it.
I did a T2 transfer of Melmak onto the plate and saw no growth for three days. I usually see growth in about 12 hours. I thought I killed it. It eventually grew, but the growth rate was incredibly slow and tomentose.
I poured more plates, with one drop of dye per media bottle. I excised a plug of Melmak from the beautiful, though dastardly plate and transferred it to the new plate. Growth appeared within 12 hours and Melmak is now flourishing. Whew!
Bottom line. If you use more than one drop of dye per media bottle, it will inhibit growth while encouraging tomentose mycelium.
Don't do it.
Are you using the food coloring that D3 recommends? I've always used 5 drops as he recommends in his tek and have had 0 issues. I recently switched to a different brand and used 7 drops with no issues. D3 has a ton of pictures of his plates while using more than 1 drop per 500mL.

Hey D3, I think I'm going to pick one of these up. I haven't been happy with the speed my alcohol lamp gets my blade red hot. One question, do you like the angled head on it? I see they also have a version that is straight vertical.

I love the angled, I feel I would prefer it over the straight up one. I have it right at the edge of my flow stream so Iโm not even outside the flow while torching. The quick one hand on and off makes for super simple operation and reduction of fuel consumption because you can just click it on and off between each sterilization reducing fire hazards.

Great! I'm convinced, just pulled the trigger on one. Seems like with the angled one you can keep the blade a bit more away from the unit too. Probably splitting hairs, but seems like a good thing.

Thereโs an air intake adjuster midway on the shaft. At first I thought mine was busted because it was like a long lighter flame and left a bunch of soot. Took me a few to notice the adjuster. Open it to full air intake and you get the nice blue torch

Iโve been testing out this new brand of food cold to see how it performs. I was a little apprehensive about the high fructose corn syrup but so far havenโt noted any negative effects.
Chefmaster - Neon Liqua-Gel Food Coloring - Fade Resistant Food Coloring - 6 Pack- 20ml Bottles - Stunning, Vivid Colors, Lightweight & Mess Free, Easy-To-Blend Formula https://www.amazon.com/dp/B07D9GDJYZ/ref=cm_sw_r_cp_api_glt_i_2426CJZ072ADMK6THRBQ?_encoding=UTF8&psc=1
The blue is a little darker than wiltons and the neon green isnt too bad. Still need to try some the other colors. The redโs are still #3 though I may try a very diluted purple and blue together and see what happens.

I just did 60 plates using your prep technique and recipe, and man do they look nice. When I was boiling the mixture I may have heated it a little to much and had a small amount of carmalization on the bottom of the pot. After pouring I sniffed a plate and the bottle and they smelt pretty strong of carmalaized LME, I've never used it before idk if that's normal0. Will this impact my cultures? They are still perfectly clear and look great.

I canโt say for sure as thatโs never happened to me but canโt imagine it will. Just be careful next time, doesnโt really require that much a boil. I heat the water to a boil. Whisk in the ingredients, hit with the stick blender for about 30 seconds or so and then remove from heat and pour into media bottles.

I've been stoned and left the agar on the stove for a couple extra minutes and the lme started to stick to the bottom a little, I just tried to scrape it up in the broth as much as reasonable, went ahead and used it and didn't see a down side.
Verum did an experiment on pcing agar at high pressure & time and debunked the 'carmilization' worry.

Can you link that because I know we have had this conversation before, but I do find that when I run LME AGAR to long or too many times in the PC before pouring It will turn a darker brown and even has sediment in it.
