Relative noob here - would you mind explaining how moisture effects 'stubborn spores'?

Sorry if this has been answered, but I noticed the recipe says 1L water, but 1 of the bottles in original recipe pic has 250 ml. 250ml X 3 = 750ml. Are you losing water during the initial boil? Should I end up with 1L in the bottles before pc'ing? I had slightly less after boiling so I compensated with a few dashes of water to each container to end up with a total of 1L agar mix before PC-ing.
For instance, I noticed Bod talking about having yr media bottle filled to 400ml and doing 20 plates. I can do barely exactly 20 with 500 ML--am I filling the plates too thickly and ending up with thick watery agar plates that condensate to hell and back?

I don’t really boil long enough to loose anything more than a negligible amount. I don’t necessarily pour an exactly even amount in each media bottle. One may be 250 another may be 350... I just eyeball split. Since All the ingredients are already mixed theres no need to be exact. 1l should be enough to pour 40 plates and still have a little left over. You may be pouring a little too thick.

I have only poured a handful of times, but I seem to get roughly 25 plates from 500ml (20ml per dish on average).

Guys,
After growing Rustywhyte and B+ on the beautiful green agar, I tried the orange dye and the charcoal. I made the T3 transfers yesterday. The myc is already happily growing.

Next time, I need to blend my charcoal better. Otherwise, I dig these pretty plates.
Thanks again D3monic!

tried this and wound up with very nice clear dishes--however:
-many plates have a few bubbles, which I don't mind but can be a little annoying
-some plates have like a big drop of standing water that rolls around when tilted--very distressing! I can't figure out what caused that? they do seem a bit soft
-i used a stainless steel straw to cut little circles, but my pours are so thick they ended up stuck inside the straw, which was a bummer--I switched back to just cuts for now, though I want to find a better tool and technique for this.

D3monic, thank you for the great tek!
Does this look ok? It's been about 6 days since transfer and I'm getting worried.

Variety, temps and agar recipe can give varying results in growth speed. That leading clear edge looks fine, It’s probably just digging in. A softer or more nutritious recipe will see that on occasion.

Thank you so much for the reply, D3. This was done at 2% agar and 2% LME because thats how I learned. Will try the amount of LME you recommend.

It is a third transfer from a Mazatapec streak plate!
Included some photos I took on Monday. The one on the purple agar is the plate these were transferred from.

I'm worried about this fuzzy growth on top, is that normal? I can tell underneath there is rhizomorphic (as shown in the last photo around 6 o'clock area to ~8:45 o'clock area) growth.

Looks like standard tomentos growth. I don’t like the fluffy myc myself. Always makes me second guess it. But from what I can see it looks healthy.
