Going back to the nutrient conversation, here the same strain one xfer on charcoal powder other on the wilton neon gel. Personally I don’t see a noticeable difference.

Its d3monic agar tek day!
One question... any thoughts on adding bleach to the sous vide water? seems like taking media bottles out of the PC and then bathing them in contam bisque while they wait to be poured.
Obv I don't want bleach in my plates, but i will probably set the sous vide a bit high and as I take each one out, wipe down with a presoasked alcohol towel and set in front of the flow hood to fully dry and cool the last 5 or 10 degrees.
I have yet to see if any moisture hides up under the rim of the top. Some lightning fast advice would be helpful.
Also... my IR thermometer is NOT sterile. I should have bought a new one that is made of smooth hard plastic vs the one from my cannabis toolkit which has a gummy/grippy coating that does not seem ideal. i am going to wrap the part i have to touch with aluminum foil for now. That should work. Thoughts?

Took fewer pics than I would have liked of the process, but here is the outcome.

thanks for the tek. everything went smoothly. Lots of condensation now, but I'm expecting that to subside by leaving them in the flow till room temp.
Not bad for a first go of it.

I don’t know if bleach would have a negative effect, I’m in the habit of wiping the exterior of my media bottle down with an iso damp paper towel. 1. To get anything that could of landed on it during transport and 2. Wipe off the moisture on the exterior. The iso makes it dry quickly.

The guy who made your tool isn't returning emails...
so plan B, eh?

The steel drinking straws you can buy at the supermarket is 8mm ID. got those, but seems like a bigass diameter. boba straws are like 1.5cm ID.
I ordered 3mm, 4mm and 5mm with the thinnest wall option. Easy enough to cut a piece of polycarbonate dowel and drill the right sized hole all the way thru it (to keep the back end open) and shove the tube inside with a touch of epoxy. or a countersunk screw or two...
So is the cutout in the tube really necessary? what's its purpose? in a punch or coring tool, i understand the purpose... for the leather discs that build up to come out of or to push it out from...and also, in the case that the back end of the tube is sealed, fo air to escape as the core sample goes in...
But really, we are just interested in cutting a clean circle which we are then picking up with a scalpel or tweezers, right?

All that’s correct, brass tubing from the hardware store might be cheaper and heats quickly. My tool is brass and open on both ends. The cut out is nice for flaming. I flame through the cutout and out the tip so the flame is going away from my hand not towards. I’m sure it would work just fine without it.

Ordered some of that black charcoal. Cant wait. I usually go purple so you can see contams clearly. But black is even better! Nice anime chick in the pic!

Grabbed these at Target this morning. They are narrower and shorter than standard drinking straws and these silicone tips make a perfect handle that will not melt. As pictured, I have them doubled up, but i am going to buy a set of thicker straws and see if the tips from those will fir OVER these tips. Thickness. Mo bettas. We will see how they handle the heat. Tonight at my home workbench I will dremmel out a port for heating away from my hand because that's a great idea, I had not thought of that angle. Thanks d3.

Awesome, you’ll love it I’m sure. I don’t really measure when I spoon it out. Just about half a spoon tip per media bottle. I think I roughed it out to around 1g powder to 100ml

Those look like they will work great. I’ll look them up and give them a try as well. How big is the siliconed area, big enough to comfortably grip?
Edit: oh I see the part about doubling them up.

seems quite adequate. 5" overall length... very spongey and grippy. pretty idea actually. you'd know better than I. $9.99, btw.
i guess you could put a third silicone tip on it but then your hand may be too close to the plate... you could hang the 3rd one half on, half off.

Great tek and comments! The leather punch works well for me. Most of the time, the agar puck stays on the plate and I use the scalpel to transfer it to the receptor plate but every now and again an puck will get stuck in the leather punch and I use the cutout and a thin lab spatula to push it out onto the receptor.

fwiw, i have done 30 or 40 transfers with the cocktail straw and not one got stuck. very little agar sticking to it... i feel like the beveled edges on the leather punch is asking for trouble.
i ordered 3mm, 4mm, 5mm, 6mm and 12mm... the bigger ones (straws vs "fillament tube") all come with those silicone tips which can be doubled up... i feel zero heat from the tool, flaming a full inch.
i am sold. i have those leather punches coming on tuesday, but i think i am going to return the set.
