Yo D3monic,

Have you noticed any differences in growth (Magenta aside) between non-colored plates and colored? I checked the Wilton box of neon color dyes and they all contain corn syrup & glycerin. I'm wondering if this alters the nutrient profile of our plates at all in any meaningful way?

The primary color set by Wilton, on the other hand, contains sorbitol & glycerin instead of corn syrup as a sweetener. Curious about that as well.

Not a big noticeable difference. Now I’ve never actually not used colored plates. But the charcoal powder plates should be free of any additional nutrients and compared to the neon gel plates there’s no notable difference between the two.

No not at all but that one does look a lil rough inside. Did you start pouring your agar thicker or anything ? Maybe tool still too hot?

Going to have to try this tek. My Petris work great but having them be more symmetrical would add to how satisfying agar work is! Thanks!

Clean all that carbonized shit off and it’ll work much better.

Hey D3,
Here's something you might get a kick out of.

Of course I'm still working in a SAB so this aid is more pronounced.
So quickly, I put a dot with a Sharpie dead center on the stack of
two plates that support the plate for inoculating.
It's easy to see the dot and is less effort to gauge dead center. Voilà
Shucks can't see it, but you get the idea...

Another buddy uses something similar but it’s more a wire mesh grid instead a cookie rack. Either way the flesh isn’t touching the print keeping it sterile other than the spores that drop.

Thanks for showing me the ropes with your guide, D3. This was my most aesthetic culture yet.

Also, I've been experimenting with Americolor's gel dyes and so far it seems promising.

https://www.amazon.com/Coloring-AmeriColor-Student-Bottles-Colors/dp/B00KCUXRLC/ref=sr_1_1?dchild=1&keywords=food+coloring+americolor+student+kit&qid=1601415142&s=home-garden&sr=1-1

Here's another example using their black dye. Both these black agar photos were taken last week.

Color holds in the PC perfectly. Mycelium didn't seem to mind the black dye, either. Although I recently did a teal-colored run and am confused about whether I just have tomentose genetics or if the dye is screwing with it. For instance, the dish below is the subsequent transfer of the culture in the first photo (T3 -> T4 with an invitro pin as T0), but it's going tomentose/fuzzy where I'd expect more rhizomorphic growth.

I dunno, what do you make of the non-rhizo shit? When this pops up with a thick translucent edge I get O_o; Maximize the img after opening for the full resolution.

Same exact thing has been happening with my teal plates. The bottom also turns brownish as if the myc is digesting the dye

At first I was worried that I had transferred mold or something, because my clone culture had not exhibited this type of growth up to that point. But to test this I did two transfers from the same plate: one to a pasty plate using magenta dye and the other to my petri using teal dye

The magenta dye looks nice and healthy while the teal one looks tomentose AF. Not sure if it is the dye or not, but I had been worried about this ever since I started using these teal plates, so thanks for posting this

The myc will consume the dye that’s pretty normal. I’ve even seen it turn pinkish on a plate that had magenta. It’s a food source just like the malt extract. I’m in the same boat as I don’t trust the wispy tomentos myc like that. Not to say it’s bad necessarily I’d take a tiny tip off the rhizo on your transfer.

Will do.

Anyone know if the shit we encounter with dyes also happens to cultures that are living in MEAC? I'm wondering if Carbon screws with the growth in the same way that dye does. Since it isn't a nutrient I'd expect that it doesn't...?
