Yeah I was thinking that it might work if the punch didn't have the hole on the side.
I keep saying D3, do something with one of those stainless steel straws. I think Yeti makes them. This is definitely a game changer!

The punch spots. These plates have been sampled. I wrapped them back up and let them grow.
Unless you mean the little satelites near the edge. It is part of the outside of the plate.
There is also some floating junk in my agar cause I didn't feel like straining it with cheese cloth.
It all gets PCd anyway.

So I started using like the 5/16 punch opting for a smaller sample size. I started out with the 3/8. I am noticing a lot more organization in the growth and less sectoring.
Will post pictures in a day or two of some Nats!

I just think it is cool that the punch is now D3monics thing! Can we get some one to add this to a sticky?
Yeah also that looks like mold and mycelium on the one plate in question. Strange I haven't seen a plate do that so clearly before.

Hermansonking said: I just did 60 plates using your prep technique and recipe, and man do they look nice. When I was boiling the mixture I may have heated it a little to much and had a small amount of carmalization on the bottom of the pot. After pouring I sniffed a plate and the bottle and they smelt pretty strong of carmalaized LME, I've never used it before idk if that's normal0. Will this impact my cultures? They are still perfectly clear and look great.
I've been stoned and left the agar on the stove for a couple extra minutes and the lme started to stick to the bottom a little, I just tried to scrape it up in the broth as much as reasonable, went ahead and used it and didn't see a down side.
Verum did an experiment on pcing agar at high pressure & time and debunked the 'carmilization' worry.
Can you link that because I know we have had this conversation before, but I do find that when I run LME AGAR to long or too many times in the PC before pouring It will turn a darker brown and even has sediment in it.

Well I am still working on this putting coloring agents in plates and taking the time to let the condensation settle infront of the FH. Still not quite where I want it but its better than the last batch.

Im still working on how much to put into each bottle. I used 1/2 Tsp and each bottle poured out to a little over 300 ML. But this is the lightest plate also.

Yeah C10, I do that when making gravy. I mix the dry ingredients with cold water and then slowly whisk into boiling water. I should try this approach when making plates. I have way to many poured plates at the moment because I was working on CHarcoal Powder levels and so I guess we wait a month?

I have to say that between your tek and C10's you guys have really upped my agar game. I'm getting nice clean plates with no sediment. The nutrient profile looks good and the color compliments my Black Sunflower seeds nicely.
